Anaphase A and B

La segregazione cromosomica avviene durante l'anafase: quando i cromatidi fratelli si separano e i singoli cromatidi si spostano verso i poli opposti della cellula.
La progressione dell'anafase costituisce due processi indipendenti ma sovrapposti: l'anafase A e l'anafase B.
Durante l'Anafase A, in assenza di coesione fratello-cromatide, due forze verso i poli agiscono sui cromosomi.
La depolimerizzazione dell'estremità positiva dei microtubuli al cinetocore produce una forza verso i poli. Il flusso di microtubuli, dalla depolimerizzazione dell'estremità negativa, genera anche una forza verso i poli. La combinazione di queste due forze verso i poli, accompagnata da un accorciamento dei cinetocorei-microtubuli, tira i singoli cromatidi verso i poli del fuso.
Quando i cromosomi figli si spostano verso i poli, inizia l'anafase B e i poli del fuso vengono separati, allungando il fuso. Le proteine motrici, la chinesina-5 e la dineina, guidano la separazione dei poli-fuso.
Le proteine motrici della chinesina-5 reticolano le estremità positive dei microtubuli interpolari sovrapposti. Queste proteine motorie dirette all'estremità positiva generano una forza all'indietro lungo i microtubuli, allontanando i poli del fuso. 
Le proteine motrici della dineina collegano le estremità dei microtubuli astrali con i componenti della corteccia cellulare. Queste proteine motorie dirette all'estremità negativa generano una forza, tirando i poli del fuso verso la corteccia cellulare.

I microtubuli si formano attraverso la polimerizzazione end-to-end degli eterodimeri di tubulina. I microtubuli del cinetocore hanno origine dai poli del fuso e le loro estremità positive si collegano con i cinetocori sui cromatidi fratelli. I complessi proteici Ndc80, presenti sul cinetocore, formano collegamenti a bassa affinità con l'estremità positiva di questi microtubuli del cinetocore.

La depolimerizzazione dell'estremità positiva rilascia eterodimeri di tubulina dalla regione terminale del microtubulo. Quando le subunità della tubulina vengono perse, i complessi Ndc80 si staccano e si riattaccano nei siti davanti al segmento di depolimerizzazione del microtubulo. Il processo si traduce in uno spostamento verso i poli, attirando il cinetocore e il cromatide associato verso il polo del fuso.

Il flusso dei microtubuli attira i cromatidi verso i poli del fuso.

Le subunità di tubulina che formano il reticolo dei microtubuli si muovono continuamente verso l'estremità negativa, esibendo un flusso di microtubuli diretto all'estremità negativa.

Il flusso dei microtubuli si sviluppa quando la depolimerizzazione continua all'estremità negativa è bilanciata dalla polimerizzazione continua all'estremità positiva. Una velocità costante di depolimerizzazione e polimerizzazione mantiene una lunghezza fissa dei microtubuli mentre le singole subunità all'interno del reticolo si muovono verso l'estremità di depolimerizzazione.

I microtubuli del cinetocoro sottoposti a flusso trascinano i cinetocori e i cromatidi associati lungo la direzione del flusso, verso i poli del fuso.

Proteine motrici dei microtubuli – Dineina e Kinesina-5

Le dineine sono proteine motorie dirette all'estremità negativa dei microtubuli. Le dineine collegano le estremità positive dei microtubuli astrali con i componenti citoscheletrici nella corteccia cellulare, posizionando così i poli del fuso all'interno della cellula. Il movimento diretto delle dineine verso l'estremità negativa genera una forza che attira i poli del fuso verso la corteccia cellulare.

Le kinesine-5 sono proteine motorie dirette all'estremità positiva. La kinesina-5 interagisce con le estremità positive dei microtubuli interpolari antiparalleli nella zona centrale del fuso. Questi motori dei microtubuli aiutano i microtubuli interpolari a scorrere uno accanto all'altro generando una forza che allontana i poli del fuso.

Anaphase is the stage of cell division when chromosomes move apart. In this phase, sister chromatids separate and travel toward opposite poles of the cell.

Chromosome movement in anaphase can happen in two ways. In anaphase A, chromosomes move closer to the poles as the microtubules attached to them shorten. In anaphase B, the spindle poles themselves move farther apart and help stretch the cell.

These two movements work together to ensure that the separated chromosomes are pulled to opposite sides. The result is a more even distribution of genetic material before the cell finishes dividing.
