Transcription

La trascrizione è il processo di sintesi dell'RNA da un modello di DNA.
Sul DNA stampo, il complesso di pre-iniziazione della trascrizione si assembla attorno al promotore principale di un gene. Il promotore include la scatola TATA e una sequenza iniziatrice. I fattori di trascrizione generali si legano alla scatola TATA, mentre la sequenza iniziatrice contiene il sito di inizio della trascrizione.
Una volta che i componenti necessari sono legati, il complesso di preiniziazione svolge un breve tratto del DNA a monte del sito di inizio della trascrizione. Successivamente, i fattori di trascrizione generali vengono dissociati dal filamento e l'RNA polimerasi inizia a produrre un nuovo filamento di mRNA.
I nucleotidi vengono aggiunti uno per uno e la sintesi dell'mRNA avviene in una direzione da cinque a tre primi, leggendo dal filamento stampo, tranne che le timidine sono sostituite dalle uridine.
Questo filamento di mRNA appena creato rappresenta una copia delle informazioni nel filamento codificante. La sintesi continuerà fino a quando non si incontrerà una sequenza di terminazione, che rilascerà l'mRNA appena prodotto.

La trascrizione è la sintesi dell'RNA da una sequenza di DNA mediante la RNA polimerasi. È il primo passo nella produzione di una proteina da una sequenza genetica. Inoltre, molte altre proteine e sequenze regolatrici sono coinvolte nella corretta sintesi dell’RNA messaggero (mRNA). La regolazione trascrizionale è responsabile della differenziazione di diversi tipi di cellule e spesso della corretta risposta cellulare ai segnali ambientali.

La trascrizione può produrre diversi tipi di molecole di RNA

Negli eucarioti, il DNA viene prima trascritto in pre-mRNA, che può essere ulteriormente trasformato in un mRNA maturo che funge da modello per la sintesi delle proteine. Tuttavia, nei procarioti, la traduzione dell'RNA in polipeptidi può iniziare mentre la trascrizione è ancora in corso. La trascrizione può anche produrre diversi tipi di molecole di RNA che non codificano per proteine, come microRNA (miRNA), RNA di trasferimento (tRNA) e RNA ribosomiale (rRNA), che influenzano tutti la sintesi proteica.

La regolazione della trascrizione è fondamentale per lo sviluppo

Con poche eccezioni, tutte le cellule del corpo umano hanno le stesse informazioni genetiche, ma diversi tipi di cellule regolano in modo differenziale la trascrizione durante lo sviluppo. Nello specifico, la regolazione trascrizionale svolge un ruolo centrale nella differenziazione cellulare, il processo di produzione di cellule specializzate, come le cellule muscolari, dalle cellule precursori meno specializzate. Alcuni geni nelle cellule precursori devono essere attivati e altri disattivati per produrre cellule specializzate.

Questo processo di differenziazione cellulare è orchestrato da proteine leganti il DNA chiamate fattori di trascrizione che controllano il livello di trascrizione dei geni. Ad esempio, durante lo sviluppo iniziale dei vertebrati, le cellule nello strato ectodermico dell'embrione in via di sviluppo ricevono diversi segnali di induzione da proteine come BMP, WNT e SHH. Questi segnali attivano fattori di trascrizione che attivano o disattivano una serie di geni. In questo modo, la regolazione trascrizionale determina se le cellule dell'ectoderma diventano cellule della pelle o cellule del sistema nervoso.
