La calibrazione dello strumento garantisce misurazioni accurate senza errori determinati.
La taratura è la regolazione del segnale misurato di uno strumento utilizzando uno standard. Ad esempio, una bilancia analitica che misura la massa deve essere tarata rispetto a un peso standard come riferimento.
In questo caso, gli errori determinati sono ancora possibili attraverso la galleggiabilità dell'aria, che rende un oggetto più leggero nell'aria che nel vuoto. Qualsiasi disparità di densità tra l'oggetto da pesare e i pesi di calibrazione viene risolta applicando una correzione al peso dell'oggetto.
La relazione tra il peso reale di un oggetto e il suo peso nell'aria considera la densità dell'oggetto, il peso di calibrazione e l'aria in condizioni standard di laboratorio.

Il processo inizia con la preparazione di una serie di soluzioni standard che, di solito, vanno da basse ad alte concentrazioni per formare un'ampia curva di calibrazione. Ogni soluzione standard viene quindi misurata per l'assorbanza utilizzando lo spettrofotometro. Lo spettrofotometro irradia la luce attraverso la soluzione e misura la quantità di luce che viene assorbita dall'analita.

I valori di assorbanza vengono tracciati rispetto alle concentrazioni note. Secondo la legge di Beer-Lambert, questo grafico dovrebbe formare una linea retta, mostrando una relazione diretta tra la concentrazione e l’assorbanza. Una volta stabilita la curva di calibrazione, la soluzione sconosciuta viene misurata per la sua assorbanza. Questo valore di assorbanza aiuta a determinare la concentrazione corrispondente dell'analita nella soluzione sconosciuta dalla curva di calibrazione.

An analytical balance measures mass and needs regular calibration to stay precise. It is calibrated by comparing its readings with known standard weights. These weights act as references for correct mass measurements. Air buoyancy can affect the balance because it can make a weighted object seem lighter than it really is in a vacuum.

A buoyancy correction is used to adjust for that error. The correction depends on the object&amp;#39;s density, the air density, and the calibration weights. By accounting for the determinate error caused by air buoyancy, the analytical balance gives a more accurate reading of an object&amp;#39;s true weight.

A spectrophotometer is calibrated in a different way because it measures how much light a solution absorbs. The process uses a calibration curve made from standard solutions with known analyte concentrations. First, a series of standards is prepared from low to high concentration to create a broad curve. Then each standard is measured for absorbance.

The spectrophotometer shines light through the solution and records how much is absorbed by the analyte. The absorbance values are plotted against the known concentrations. According to the Beer-Lambert law, the plot should be a straight line, showing a direct link between concentration and absorbance. After the curve is made, an unknown solution can be measured, and its absorbance is used to find the analyte concentration from the curve.
