La preparazione del campione è una fase fondamentale per ottenere risultati di analisi accurati. Comprende tre obiettivi principali: estrazione, purificazione e concentrazione.
In primo luogo, l'estrazione consiste nell'isolare una miscela con l'analita di interesse da una matrice di campioni. In secondo luogo, se sono presenti sostanze interferenti, queste vengono rimosse per ottenere un campione puro. In terzo luogo, se l'analita estratto è a bassa concentrazione, viene concentrato a un livello che può essere rilevato con precisione.
Le forme comuni del campione sono solido, liquido e gassoso. I pezzi solidi troppo grandi per essere analizzati direttamente vengono polverizzati con metodi fisici come la macinazione, la frantumazione e la macinazione.
La setacciatura e la miscelazione di questa miscela si ottengono una polvere fine omogenea, che viene poi essiccata e disciolta in un solvente adatto per estrarre l'analita nella soluzione. 
A differenza dei solidi, le sospensioni liquide vengono filtrate o centrifugate prima dell'analisi per rimuovere le particelle solide impure.
Poiché gli assorbenti solidi adsorbono selettivamente l'analita gassoso, non è necessaria la preparazione del campione per i gas.

La preparazione del campione è una fase essenziale del processo analitico. Comporta la preparazione di un campione in modo che possa essere analizzato in modo accurato. L'obiettivo è estrarre l'analita, la sostanza che si desidera misurare, dal campione, rimuovendo al contempo tutti i componenti che potrebbero interferire con l'analisi. Le tecniche di preparazione del campione variano a seconda dello stato fisico di quest’ultimo.

I campioni solidi sfusi, o di grandi dimensioni, di solito vengono ridotti di dimensioni utilizzando tecniche di macinazione, frantumazione o fresatura per aumentare la superficie del campione e fornire un'analisi più efficiente. Dopo la macinazione, il campione viene setacciato e miscelato per garantire una composizione uniforme. Il campione, una volta in polvere, viene quindi sciolto in un solvente adatto per estrarre l'analita dalla matrice solida. La scelta del solvente dipende dalla natura dell'analita e dalle caratteristiche di solubilità dei componenti del campione.

I campioni liquidi che contengono le particelle solide sospese, o i precipitati, sono convenzionalmente sottoposti a filtrazione, o centrifugazione, per separare le impurità solide dal liquido. La filtrazione comporta il passaggio del liquido attraverso un filtro, mentre la centrifugazione usa la forza centrifuga (rotazione) per separare i solidi dalla fase liquida. Se l'analita è presente in tracce, i campioni liquidi vengono sottoposti a pretrattamento di fasi di concentrazione per aumentare la concentrazione dell'analita per una migliore rilevazione.

La preparazione del campione, solitamente, è superflua per l'analisi di analiti gassosi perché il campione di gas può essere introdotto direttamente nello strumento analitico.

Sample preparation depends on the state and form of the sample before analysis. It helps make the sample ready for accurate measurement. The main goal is to extract the analyte, which is the substance being measured, and remove parts that could interfere with the analysis.

Solid samples often need the most handling. Large solids are usually ground, crushed, or milled to make them smaller and increase surface area. The sample is then sieved and mixed so it has a uniform composition. After that, the powdered material is dissolved in a suitable solvent to pull the analyte out of the solid matrix. The best solvent depends on the analyte and on how well the sample components dissolve.

Liquid samples may also need treatment before analysis. If they contain suspended solids or precipitates, they are usually filtered or centrifuged to remove the unwanted particles. Filtration works by passing the liquid through a filter. Centrifugation uses spinning force to separate the solid material from the liquid phase.

When the analyte is present in trace amounts, liquid samples may need a concentration step. This increases the analyte level so it is easier to detect. Sample preparation is usually not needed for gaseous analytes because the gas sample can be introduced directly into the analytical instrument.
