Tipicamente, a basse concentrazioni dell'analita, l'intensità dell'emissione è linearmente proporzionale alla concentrazione dell'analita nella soluzione.
Tuttavia, a concentrazioni più elevate dell'analita, si osserva una deviazione da questa relazione lineare, principalmente a causa dell'aumento dell'autoassorbimento della radiazione emessa dagli atomi dello stato fondamentale.
L'applicazione della fotometria a fiamma include la determinazione quantitativa di metalli alcalini e alcalino-terrosi, principalmente in campioni clinici, di biodiesel e di cemento.

In un fotometro a fiamma, quando una soluzione come il cloruro di potassio viene aspirata nella fiamma, il solvente evapora, lasciando dietro di sé sale disidratato. Questo sale si dissocia in atomi gassosi liberi nel loro stato fondamentale. Alcuni di questi atomi assorbono energia dalla fiamma, portando alla loro eccitazione. Gli atomi eccitati tornano allo stato fondamentale, emettendo dei fotoni a lunghezze d'onda caratteristiche. Siccome sono coinvolte solo transizioni elettroniche, le linee di emissione risultanti sono molto strette. L'intensità della radiazione emessa è linearmente proporzionale alla concentrazione dell'analita nella soluzione aspirata. Tuttavia, questa linearità si osserva principalmente all'estremità inferiore della curva di calibrazione. A concentrazioni di analita più elevate, più atomi nello stato fondamentale, che possono riassorbire la radiazione emessa, si discostano dalla relazione lineare.

I moderni fotometri a fiamma possono elaborare automaticamente i dati di calibrazione multianalita multipunto ed eseguire misurazioni senza la necessità di diluizione del campione, adattandosi a concentrazioni fino a 1000 mg/L. Mentre la fotometria a fiamma viene principalmente usata per la determinazione di sodio (Na), potassio (K) e litio (Li) nei laboratori clinici, trova anche applicazioni di nicchia nei vari campi. Per esempio, misura i metalli alcalini residui nel biodiesel e determina il sodio, il potassio e il calcio nel cemento. In passato, la spettrometria a emissione di fiamma consentiva la misurazione di un massimo di 60 elementi in fiamme calde di protossido di azoto e acetilene. Tuttavia, oggi, la spettrometria ad assorbimento atomico, o AAS, è usata prevalentemente per misurare metalli diversi dai metalli alcalini.

Flame photometry is a method for detecting metals by measuring the light they emit in a flame. A solution such as potassium chloride is aspirated into the flame. The solvent evaporates first. The remaining salt becomes dehydrated and then dissociates into free gaseous atoms in the ground state.

Some of these atoms absorb energy from the flame and move to an excited state. When they return to the ground state, they emit photons at characteristic wavelengths. These emissions are very narrow because only electronic transitions are involved. The light intensity increases with analyte concentration in the sample.

This relationship is mainly linear at the low end of the calibration curve. At higher concentrations, the line can bend because more ground-state atoms can reabsorb the emitted radiation. Modern flame photometers can handle multianalyte, multipoint calibration data automatically. They can also measure samples without dilution and can accept concentrations up to 1000 mg/L.

Flame photometry is used most often for sodium, potassium, and lithium in clinical laboratories. It also has niche uses in other areas. For example, it can measure residual alkali metals in biodiesel and determine sodium, potassium, and calcium in cement. Flame emission spectrometry once measured up to 60 elements in hot nitrous oxide and acetylene flames, but atomic absorption spectrometry, or AAS, is now mainly used for metals other than alkali metals.
