Peg Crosslinkers

PEG crosslinkers are polyethylene glycol-based molecules that join polymer chains to form interconnected networks, making them valuable tools for engineering biomaterials with tunable structure and properties. Their reactive end groups form covalent bonds with complementary functional groups on PEG or other polymers, converting liquid polymer solutions into stable three-dimensional hydrogels under controlled chemical or biological conditions. In bioengineering, these crosslinked materials can provide hydrated, tissue-like environments for cell culture, drug delivery, wound treatment, and regenerative medicine. By adjusting crosslinker architecture, concentration, and reaction conditions, researchers can control gel stiffness, swelling, degradation, and transport of biomolecules.

Poly(ethylene glycol) (PEG) brush-arm star polymers (BASPs) with narrow mass distributions and tunable nanoscopic sizes are synthesized in via ring opening metathesis polymerization (ROMP) of a PEG-norbornene macromonomer followed by transfer of portions of the resulting living brush initiator to vials containing varied amounts of a rigid, photo-cleavable bis-norbornene crosslinker.

This video demonstrates the crosslinking of neuronal surface proteins using Bis-sulfosuccinimidyl suberate or BS3. Isolated and chilled mouse brain is sectioned coronally. The desired region is extracted from the sections, minced, and treated with BS3. BS3 crosslinks surface proteins like GABA receptors, leaving internal proteins unaltered. The reaction is quenched with glycine before processing the samples for further analysis.
