Ex Vivo Technique

Ex vivo technique refers to the study or manipulation of cells, tissues, or organs after their removal from a living organism while preserving their biological function under controlled laboratory conditions. Researchers isolate the target material and maintain it in a suitable environment, using regulated temperature, nutrients, gases, and culture media to support viability while applying defined treatments or measurements. This approach bridges in vivo biology and in vitro experimentation, enabling direct analysis of tissue responses, disease mechanisms, drug effects, and therapeutic strategies with greater experimental control. Ex vivo models also support tissue engineering, transplantation research, and personalized medicine when samples retain features of the original organism.

A Technique to Assess the In Vivo Localization of Tumor-Specific Antibodies

2025

This video demonstrates a technique to study the in vivo localization of tumor-specific antibodies in a mouse tumor xenograft model. Upon subcutaneously injecting cancer cells into mice for tumor formation, fluorescently-labeled antibodies are injected that bind to tumor-specific receptors, enabling in vivo assessment of antibody localization via imaging.

An Ex Vivo Technique to Manufacture Chimeric Antigen Receptor T Cells

2025

This video demonstrates a technique to manufacture chimeric antigen receptor (CAR) T cells ex vivo. Human T cells are incubated with magnetic beads coated with anti-CD3 and anti-CD28 antibodies, along with a medium supplemented with interleukin-2 (IL-2), leading to T cell activation and proliferation. Transgenic lentiviral vectors are introduced, allowing the transgenic RNA to integrate into the host genome and express the transgene. This results in the transduced T cells expressing chimeric...

Ex vivo Lung Perfusion: A Technique for Pulmonary Research in a Rabbit Model

This video shows an ex vivo technique for isolated lung perfusion. It is a tool for pulmonary research that will aid studies of various physiological and pathological mechanisms involved in lung transplantation procedures.

An In Vivo Technique to Track Bispecific Antibody-Induced T Cell Trafficking to Tumors

2025

This video demonstrates a method for tracking bispecific antibody-induced T cell trafficking to tumors in a mouse model. Luciferase-transduced effector T cells are injected into a mouse with a subcutaneous tumor cell xenograft. Bispecific antibodies are then introduced, binding to T cells and the tumor cells. The localization of the T cells is tracked using bioluminescent imaging, with the magnitude of the signal indicating the extent of T cell infiltration into the tumor.
