Embryo Grafting Technique

Embryo grafting is a biological technique in which an isolated plant embryo is attached to a compatible living rootstock so it can continue developing, making the method valuable for studying and rescuing embryos that cannot mature independently. Typically, researchers excise the embryo under sterile conditions and place it onto a prepared seedling or rootstock, where the graft establishes functional contact and supplies water and nutrients during development. The technique supports plant breeding by recovering embryos from wide or otherwise difficult crosses, accelerating the production of viable plants, and enabling investigation of embryo development, graft compatibility, and interactions between developing tissues.

A Technique for Human Skin Grafting in a Mouse Model

This video demonstrates a method for grafting human skin onto an immunodeficient mouse with a suppressed neutrophil response. The method involves applying human split-thickness skin to the mouse's graft bed, sealing it, and covering it for recovery. Eventually, the human skin adheres to the mouse's skin, establishing vascularization and circulation, resulting in a successful skin xenograft.

By grafting beads soaked in growth factors or specific inhibitors of signaling pathways into developing embryos it is possible to directly test their effects in vivo. In this protocol beads are grafted into the limb bud to determine the effects of these molecules on gene expression and signal transduction.

Here we report dual labeling of neural crest cells and blood vessels using chickGFP neural tube intraspecies grafting combined with intra-vascular DiI injection. This experimental technique allows us to simultaneously visualize and study development of the NCC-derived (enteric) nervous system and the vascular system, during organogenesis.
