Pdgfr Alpha Fibroblasts

PDGFRα fibroblasts are connective-tissue cells identified by expression of platelet-derived growth factor receptor alpha (PDGFRα), a receptor that marks diverse fibroblast populations involved in tissue organization and repair. When PDGF ligands bind PDGFRα, the receptor activates intracellular tyrosine kinase signaling that can promote fibroblast proliferation, migration, survival, and extracellular matrix production. These cells contribute to tissue development, wound healing, and remodeling, but their dysregulation can promote pathological fibrosis. Identifying and studying PDGFRα fibroblasts helps researchers map fibroblast heterogeneity, trace cell behavior in vivo, and investigate potential targets for regenerative medicine and anti-fibrotic therapies.

Experimental Generation of Carcinoma-Associated Fibroblasts (CAFs) from Human Mammary Fibroblasts

Carcinoma-associated fibroblasts (CAFs) rich in myofibroblasts present within the tumour stroma, play a major role in driving tumour progression. We developed a coimplantation tumour xengraft model for experimentally generating CAFs from human mammary fibroblasts. The protocol describes how to establish CAF myofibroblasts that acquire an ability to promote tumourigenesis.

Isolation of Rat Portal Fibroblasts by In situ Liver Perfusion

A technique for isolating portal fibroblasts from rat liver is described. Livers are perfused and digested in situ with collagenase, followed by ex vivo digestion of the liver slurry and size selection of cells. This method provides a pure population of portal fibroblasts without the need for passage in culture.

This video demonstrates the immunostaining of neurons treated with alpha-synuclein (αS) aggregates. The neurons are first treated with αS aggregates. They are then fixed, permeabilized, and treated with a blocking solution. Primary antibodies are added, followed by a fluorescently labeled secondary antibodies and a nuclear stain. The sample is then mounted and observed under a microscope.
