Phosphatidylserine

Phosphatidylserine is an anionic glycerophospholipid found primarily on the inner leaflet of eukaryotic cell membranes, where it contributes to membrane structure and cellular signaling. Cells maintain this asymmetrical distribution through lipid transport enzymes, but during apoptosis, phosphatidylserine is exposed on the outer membrane surface, creating an “eat-me” signal that helps phagocytes recognize and remove dying cells. Its redistribution also supports blood coagulation by providing a negatively charged surface for enzyme complexes. Studying phosphatidylserine therefore helps researchers investigate membrane organization, programmed cell death, immune clearance, hemostasis, and related biological disorders.

Annexin V Binding Assay: A Fluorescence-Based Technique to Identify Apoptotic Erythrocytes via Phosphatidylserine Labeling

2025

In this video, we demonstrate the Annexin V binding assay to measure the degree of eryptosis, erythrocyte programmed cell death, using fluorescent Annexin V staining. Apoptotic erythrocytes demonstrate phosphatidylserine translocation from the inner to the outer leaflet of the cell membrane, causing the fluorophore-tagged Annexin V to bind to the translocated phosphatidylserine, thereby emitting a fluorescent signal.

Procoagulant platelet formation has been correlated with an increased risk of thrombosis. Presented here is a precise protocol for isolating washed platelets from human blood, intended to quantify the exposure of phosphatidylserine and microvesicle release, which are distinctive features of procoagulant platelets.

In Vitro Phagocytosis Assay: A Live Imaging Based Method to Study Real-time Astrocyte Mediated Synaptosome Phagocytosis

In this video, we evaluate the phagocytic capacity of purified astrocytes in vitro by live imaging assay. The assay serves as an effective screening platform to identify chemical compounds that can enhance or inhibit the phagocytic capacity of astrocytes.

An In Vitro Assay to Assess CAR T Cell Cytotoxicity against Tumor Spheroids

This video demonstrates an in vitro assay evaluating the cytotoxic effects of CAR T cells on 3D tumor spheroids expressing fluorescent proteins. This assessment utilizes the Annexin V red fluorescent probe, which binds to exposed phosphatidylserine residues on the outer leaflet of apoptotic cancer cells after interactions with CAR T cells. The red fluorescence emitted by the probe aids in distinguishing these cells from healthy ones emitting green fluorescence.
