Scaffold Protein Assembly

Scaffold protein assembly is the formation of organized protein complexes in which scaffold proteins bring selected molecular partners together, enabling efficient and coordinated cellular signaling. Through modular interaction domains, short binding motifs, multivalent contacts, and regulatory modifications such as phosphorylation, scaffolds recruit, position, and sometimes stabilize enzymes, receptors, and adaptor proteins within defined cellular compartments. This spatial organization can control signal strength, timing, and pathway specificity, while defects in assembly may disrupt processes including cell polarity, cytoskeletal dynamics, and responses to extracellular cues. Studying scaffold protein assembly helps explain how cells integrate information and supports research into disease mechanisms, synthetic signaling systems, and abnormal protein interactions.

A method to obtain nanofibers and complex nanostructures from single or multiple extracellular matrix proteins is described. This method uses protein-surface interactions to create free-standing protein-based materials with tunable composition and architecture for use in a variety of tissue engineering and biotechnology applications.
