Vimentin Fibroblast Marker

Vimentin is an intermediate filament protein commonly used as a fibroblast marker because fibroblasts typically express this structural component of the mesenchymal cytoskeleton. In immunofluorescence or immunohistochemistry, antibodies bind vimentin within cells, producing a detectable signal that reveals filament organization and helps identify fibroblast-rich populations. Vimentin staining supports the study of cell morphology, wound repair, tissue remodeling, and fibrosis in cultured cells and tissue samples. Because vimentin is also expressed by many other mesenchymal and non-epithelial cells, researchers generally combine it with additional morphological or molecular markers to confirm fibroblast identity and distinguish related cell types.

Isolation of CD 90+ Fibroblast/Myofibroblasts from Human Frozen Gastrointestinal Specimens

Here, a protocol to isolate and establish primary fibroblast/myofibroblast (MF) cultures from frozen gastric, small intestinal, and colonic tissue-yielding cells with a MF phenotype-is presented. These cells express CD90, α-SMA and vimentin. MFs can be used for a variety of functional assays including enzymatic activity and cytokine production.

Carcinoma-associated fibroblasts (CAFs) rich in myofibroblasts present within the tumour stroma, play a major role in driving tumour progression. We developed a coimplantation tumour xengraft model for experimentally generating CAFs from human mammary fibroblasts. The protocol describes how to establish CAF myofibroblasts that acquire an ability to promote tumourigenesis.

Generating Primary Fibroblast Cultures from Mouse Ear and Tail Tissues

We describe a simple and quick experimental procedure for generating primary fibroblasts from the ears and tails of mice. The procedure does not require special animal training and can be used for the generation of fibroblast cultures from ears stored at RT for up to 10 days.
