Jones Test

The Jones test is a qualitative oxidation test used in organic chemistry to identify primary and secondary alcohols and distinguish them from tertiary alcohols. It uses chromic acid, typically generated from chromium trioxide and sulfuric acid in acetone, to oxidize primary alcohols to carboxylic acids and secondary alcohols to ketones; reduction of orange chromium(VI) to green chromium(III) produces the characteristic positive color change. Tertiary alcohols generally do not react under these conditions, so the test helps classify alcohol functional groups. Although instrumental methods provide greater specificity, the Jones test remains a rapid teaching and laboratory screening method for assessing oxidation behavior.

The Vermicelli and Capellini Handling Tests of forepaw dexterity take advantage of the natural inclination of rodents to manipulate food items using skillful forepaw and digit movements. Animals are videotaped while handling short strands of uncooked dry pasta. Slow motion video playback allows for the quantification of forepaw adjustments.

This paper describes a novel method for oral infection of mice using Listeria monocytogenes-contaminated food. The protocol can readily be adapted for use with other food borne bacterial pathogens.

The cryosphere offers access to preserved organisms that persisted under past environmental conditions. A protocol is presented to collect and decontaminate permafrost cores of soils and ice. The absence of exogenous colonies and DNA suggest that microorganisms detected represent the material, rather than contamination from drilling or processing.
