Peptide Characterization

Peptide characterization is the analytical process of determining a peptide’s identity, sequence, composition, purity, and structural features. In chemistry, researchers commonly separate peptides by liquid chromatography and measure their molecular masses with mass spectrometry; tandem fragmentation patterns can then support sequence assignment, while spectroscopic methods provide complementary information about conformation and chemical environment. These measurements help detect truncations, oxidation, deamidation, aggregation, and other modifications that influence peptide behavior. Characterization is therefore essential for confirming synthetic products, assessing stability and batch consistency, and evaluating peptide-based probes, therapeutics, and biomaterials in research and development.

2014

Peptide tertiary amides (PTAs) are a superfamily of peptidomimetics that include but are not limited to peptides, peptoids and N-methylated peptides. Here we describe a synthetic method which combines both split-and-pool and sub-monomer strategies to synthesize a one-bead one-compound library of PTAs.

This paper describes the formation of highly ordered peptide-based structures by the spontaneous process of self-assembly. The method utilizes commercially available peptides and common lab equipment. This technique can be applied to a large variety of peptides and may lead to the discovery of new peptide-based assemblies.
