Acetaminophen Overdose

Acetaminophen overdose is a toxic exposure that occurs when the body receives more acetaminophen than the liver can safely process, making it a major cause of acute liver injury. At therapeutic doses, the liver primarily converts acetaminophen into harmless metabolites, but excessive amounts increase production of the reactive metabolite NAPQI; when glutathione stores are depleted, NAPQI damages hepatocytes and can cause hepatic failure. Clinical evaluation uses dose history, timing, serum acetaminophen concentrations, and liver-function tests to assess risk. Prompt treatment with the antidote N-acetylcysteine replenishes glutathione and reduces liver injury, emphasizing the importance of early recognition and medication safety.

The acute liver failure animal model developed in the current study presents a feasible alternative for the study of potential therapies. The current model employs the combined effect of physical and drug-induced hepatic injury and provides a suitable time window to study the potential of novel therapies.

An optical system was developed to visualize hepatic microcirculation with FITC-labeled erythrocytes and to measure the partial pressure of oxygen in the microvessels with laser-assisted phosphorimetry. This method can be used to investigate physiological and pathological mechanisms by analyzing microvascular structure, diameter, blood flow velocity, and oxygen tension.
