Macro Protocol

A macro protocol is a coordinated framework that combines multiple laboratory procedures into one standardized workflow, helping researchers perform complex experiments consistently and efficiently. In genetics, it links sequential steps such as sample preparation, nucleic acid handling, amplification, separation, and result analysis, with defined conditions and checkpoints guiding the transition between stages. By organizing these interdependent procedures, macro protocols reduce variation, clarify experimental timing, and support reproducible data collection. They are useful for teaching multi-step genetic methods, documenting research workflows, and adapting established procedures for applications such as genotyping, DNA analysis, and molecular diagnostics.

We describe the fabrication of micropatterned hydrogel sheets using a simple process, which can be assembled and manipulated in a freestanding form. Using these modular hydrogel sheets, a simple macro-scaled 3D cell culture system can be generated with a controlled cellular microenvironment.
