Mutant Strain Generation

Mutant strain generation is the deliberate production of organisms with altered genetic material to investigate gene function, biological pathways, or useful traits. In genetics, researchers introduce or select mutations through methods such as targeted gene modification, random mutagenesis, or transfer of altered genetic sequences, then isolate candidate strains and confirm the intended genotype and phenotype. These strains provide controlled systems for linking DNA changes to observable characteristics, including growth, metabolism, development, or resistance. Mutant strain generation supports functional genomics, disease modeling, microbial biotechnology, and studies of heredity, while enabling researchers to test how specific genetic changes affect biological systems.

Introducing multiple genomic alterations into cyanobacteria is an essential tool in the development of strains for industrial and basic research purposes. We describe a system for generating unmarked mutants in the model cyanobacterial species Synechocystis sp. PCC6803 and marked mutants in Synechococcus sp. PCC7002.

Source: Lea-Smith, D. J., et al. Generation of Marked and Markerless Mutants in Model Cyanobacterial Species. J. Vis. Exp. (2016)The video demonstrates a method for generating antibiotic-marked mutants in cyanobacteria. The protocol begins with culturing cyanobacteria, followed by centrifugation and washing steps that preserve surface pili. A plasmid carrying an antibiotic resistance gene flanked by homologous sequences is then introduced. During incubation, the plasmid is internalized via pili...
