Streptavidin Hrp

Streptavidin HRP is a molecular detection reagent that combines streptavidin, a protein with exceptionally strong affinity for biotin, with horseradish peroxidase (HRP), an enzyme that produces a measurable signal. In genetic assays, streptavidin binds biotinylated DNA, RNA, primers, or probes, concentrating the HRP component at the target site; HRP then catalyzes oxidation of a chromogenic or chemiluminescent substrate in the presence of hydrogen peroxide. This signal enables sensitive visualization and quantification of nucleic acids in techniques such as Southern blotting, hybridization assays, and microarray analysis. Streptavidin HRP therefore links biotin-based molecular recognition to enzyme-amplified detection in genetics research.

Source: Pohl, M. O. & Stertz, S. Measuring Attachment and Internalization of Influenza A Virus in A549 Cells by Flow Cytometry. J. Vis. Exp. (2015)This video demonstrates the use of a streptavidin blocking assay to distinguish surface-bound from internalized influenza virus in human lung epithelial cells. Comparing fluorescence signals shows that a higher intensity after incubation indicates successful viral internalization.
