Anti-infective Evaluation

Anti-infective evaluation is the systematic assessment of compounds, therapies, or immune-based interventions for their ability to prevent, control, or eliminate infectious disease. In laboratory studies, researchers expose target microorganisms or infected cells to a candidate treatment and measure outcomes such as microbial growth, viability, replication, or host-cell responses under defined conditions. These evaluations can compare antimicrobial potency, selectivity, and resistance development while clarifying how an intervention interacts with pathogens and the immune system. The results guide drug discovery, therapeutic selection, vaccine and biologic development, and investigation of host-pathogen interactions, helping translate experimental findings into safer, more effective infection-control strategies.

An Assay for Quantifying Antibody-Mediated Phagocytosis of Parasite-Infected Erythrocytes

2025

This video demonstrates a flow cytometry-based phagocytosis assay involving labeled and opsonized Plasmodium falciparum-infected erythrocytes by monocytes. Flow cytometry identifies fluorescence in monocytes, confirming the successful identification of opsonized erythrocytes through phagocytosis.

An In Vitro Assay to Examine the Antibody-Dependent Enhancement of Viral Infection

2025

This video demonstrates an assay to measure antibody-dependent enhancement (ADE) of Dengue virus infection using Zika Virus Convalescent Serum. The assay uses Dengue reporter viral particles (RVPs) encoding a green fluorescent protein. Serum antibodies against the Zika Virus cross-react with RVPs to form a complex. The bound antibodies enhance RVP uptake by cells expressing the Fc receptor, which is detected using flow cytometry.

Detection of SARS-CoV-2 Neutralizing Antibodies using High-Throughput Fluorescent Imaging of Pseudovirus Infection

The protocol described here outlines a fast and effective method for measuring neutralizing antibodies against the SARS-CoV-2 spike protein by evaluating the ability of convalescent serum samples to inhibit infection by an enhanced green fluorescent protein-labeled vesicular stomatitis virus pseudotyped with spike glycoprotein.

Evaluating the Intracellular Distribution of Virus-Derived Peptide-Modified Antibody Conjugates

2025

This video demonstrates the process of assessing the intracellular distribution of virus-derived peptide-modified antibody conjugates using confocal microscopy. The viral-derived nuclear localization sequence directs antibody-conjugates within the cytoplasm, confirmed through immunostaining and fluorescence measurement.

An In Vitro Assay to Evaluate the Effect of Antibodies on Candida tropicalis Biofilm

2025

This video demonstrates an in vitro technique to study the effect of antibodies on fungal biofilms. The pathogenic fungus Candida tropicalis secretes a virulence factor secreted aspartyl proteinase 2 (Sap2), facilitating biofilm formation. Upon introducing a heat-inactivated serum containing Sap2-specific antibodies that inhibit biofilm maturation, the viability of the fungal cells is assessed using a chromogenic assay.
