Heat-inactivated Complement

Heat-inactivated complement is serum or plasma complement whose enzymatic activity has been deliberately destroyed by controlled heating, while many other serum components, including antibodies, remain functional. Heating denatures complement proteins and interrupts the proteolytic cascade that normally generates opsonins, inflammatory mediators, and membrane attack complexes, preventing complement-dependent lysis and amplification of immune responses. In immunology and infection research, researchers use heat-inactivated serum to separate antibody-mediated effects from complement-mediated effects, prepare cell-culture supplements with reduced innate immune activity, and test whether pathogens or immune reactions depend on an intact complement system.

A Serum Bactericidal Assay for the Complement-Mediated Bactericidal Activity of Antibodies

This video demonstrates a technique to evaluate the complement-mediated bactericidal activity of antibodies present in serum. This approach combines isolated serum with pathogenic bacteria and exogenous complement proteins. Upon incubation, the bacteria are grown on an agar plate, and the produced colonies are quantified to measure the serum bactericidal titer.
