Pseudovirus Assay

Pseudovirus assays are laboratory tests that use engineered, replication-defective viral particles to measure virus entry or antibody neutralization without producing a complete infectious virus. In a typical assay, pseudoviruses display a selected pathogen surface protein and carry a reporter gene; after binding and entering susceptible cells, reporter expression provides a quantifiable signal that reflects entry efficiency, while reduced signal indicates neutralization. In immunology and infection research, these assays support analysis of receptor usage, vaccine-elicited antibodies, sera, and entry inhibitors. Their standardized, adaptable format can improve biosafety and enable comparative studies of infection-related mechanisms and immune protection.

(2021).This video demonstrates a luciferase-based assay to quantify pseudovirus infection in epithelial cells. The pseudovirus encodes the luciferase gene, which is expressed in infected cells. After washing, and cells are incubated with lysis buffer at an ultra-low temperature, and then thawed to induce lysis.

(2021)This video describes the virus plaque assay using coronavirus pseudovirus particles. The diluted pseudovirus is added to a monolayer of mammalian cells, followed by incubation with a methylcellulose overlay. Plaques formed by infected cells are counted visually to determine the infectious virus titer.
