Ldh Assay

An LDH assay is a laboratory test that measures lactate dehydrogenase activity or the amount of this enzyme released into a biological sample, helping assess cellular injury and tissue damage. In a common enzymatic format, LDH catalyzes the reversible conversion of pyruvate and NADH to lactate and NAD+, and the reaction is quantified by monitoring the change in NADH absorbance at 340 nm. In medicine, LDH assays support evaluation of hemolysis, liver and muscle injury, disease progression, and treatment response, while cell-based versions measure membrane damage and cytotoxicity in experimental research.

A Lactate Dehydrogenase Release Assay To Detect Macrophage Death Following Nigericin Exposure

2025

This video demonstrates the lactate dehydrogenase assay to detect inflammasome-mediated macrophage death. The macrophages are exposed to lipopolysaccharide for priming, followed by exposure to nigericin to cause inflammasome-induced cell death, which is determined by the lactate dehydrogenase assay.

A Lactate Dehydrogenase Release Assay to Detect Cryptococcus neoformans Infection in Macrophages

2025

In this video, we demonstrate a cytotoxicity assay to assess Cryptococcus neoformans, a pathogenic fungus, infection in macrophages. The cytotoxicity is determined by the release of cytoplasmic lactate dehydrogenase enzyme due to macrophage lysis caused by Cryptococcus neoformans infection.

An In Vitro Assay for Evaluating Natural Killer Cell Cytotoxicity against Cancer Cells

2025

This video demonstrates performing a lactate dehydrogenase release assay to determine the cytotoxicity of natural killer (NK) cells against the target cancer cells. The assay quantifies the release of the cytoplasmic lactate dehydrogenase enzyme from cancer cells by measuring the absorbance of the red-colored product formed.

Evaluating Cytotoxicity and Membrane Integrity in Lung Slices via Lactate Dehydrogenase Assay
