Alkaline Silver Solution

Alkaline silver solution is a silver-ion reagent prepared under basic conditions for selectively visualizing structures in biological tissue, including neuronal elements. In histological staining, tissue-associated compounds or preformed reaction products act as nucleation sites where silver ions are reduced to visible metallic silver deposits, producing contrast that can be examined by light microscopy. In neuroscience, this approach supports the study of neuronal morphology, axons, dendrites, and tissue changes associated with degeneration or injury. By converting otherwise difficult-to-see molecular or structural differences into localized dark deposits, alkaline silver methods help researchers map neural organization and assess pathological alterations in preserved brain sections.

The alkaline comet assay measures DNA strand breaks in eukaryotic cells. By adding an Endonuclease III or human 8-oxoguanine-DNA-N-glycosylase digestion step, the assay can efficiently detect oxidative DNA damage. We describe methods for using these assays to detect DNA damage in rat liver.
