簡易染色では、単一の色素を使用して、細胞のサイズ、形状、および基本的な特徴を強調します。

固定は、染色前に細胞の形態を維持し、構造を固定するための重要なステップです。

熱固定は、光学顕微鏡の細菌および古細菌のサンプルに一般的に使用されます。

簡単な染色を行うには、微生物培養物の薄い塗抹標本をスライド上に広げ、風乾し、熱固定します。

次に、スライドをクリスタルバイオレット、メチレンブルー、サフラニンなどの塩基性色素で1〜2分間染色します。

染色後、スライドを水ですすいで余分な染料を取り除き、吸収紙で吸水して乾かします。
