NF-κBタンパク質は、免疫細胞の活性化、細胞接着、抗菌応答、細胞周期調節など、多くの重要な細胞機能に関与する転写因子です。
静止状態では、NF-κBはIκBɑと呼ばれる抑制性タンパク質に直接結合したヘテロ二量体として存在します。IκBɑは、転写活性化因子としてのNF-κBの活性を抑制します。
NF-κB依存性シグナル伝達経路は、適切なリガンドが標的細胞上の対応する細胞表面受容体に結合すると活性化されます。
例えば、この経路は、腫瘍壊死因子-ɑのTNF受容体への結合、インターロイキン1のIL1受容体への結合、または病原体のToll様受容体への結合によって引き起こされ得る。
