Electrochemistry Fluorescence Microscopy

Electrochemistry fluorescence microscopy is a technique that combines electrochemical measurements with fluorescence imaging to visualize chemical and biological events at electrode interfaces. An applied potential drives oxidation-reduction reactions, while fluorescent probes or labels report changes in molecular concentration, local environment, activity, or state through variations in intensity, spectrum, or lifetime. Correlating spatially resolved fluorescence with current and potential reveals heterogeneous reaction kinetics, transport, and interfacial behavior that bulk measurements may obscure. In biochemistry, this approach supports studies of redox enzymes, biomolecular interactions, membrane processes, and biosensor performance by linking molecular signals to quantitative electrochemical responses.

Here, we present a protocol to study the molecular mechanism of proton translocation across lipid membranes of single liposomes, using cytochrome bo3 as an example. Combining electrochemistry and fluorescence microscopy, pH changes in the lumen of single vesicles, containing single or multiple enzyme, can be detected and analyzed individually.

Introduction to Fluorescence Microscopy

2023

Fluorescence microscopy is a very powerful analytical tool that combines the magnifying properties of light microscopy with visualization of fluorescence. Fluorescence is a phenomenon that involves absorbance and emission of a small range of light wavelengths by a fluorescent molecule known as a fluorophore. Fluorescence microscopy is accomplished in conjunction with the basic light microscope by the addition of a powerful light source, specialized filters, and a means of fluorescently...

Electrochemistry: Overview

2024

Electrochemistry is the branch of chemistry that studies the relationship between electrical quantities and chemical reactions, particularly oxidation and reduction. Oxidation is the loss of electrons from a substance, whereas reduction refers to the gain of electrons. A substance with a strong electron affinity is called an oxidizing agent (oxidant), and a reducing agent (reductant) is a species that donates electrons. Oxidation and reduction processes are pivotal to electrochemical reactions,...

A lensless on-chip fluorescent microscopy platform is demonstrated that can image fluorescent objects over an ultra-wide field-of-view of e.g., >0.6-8 cm2 with <4μm resolution using a compressive sampling based decoding algorithm. Such a compact and wide-field fluorescent on-chip imaging modality could be valuable for high-throughput cytometry, rare-cell research and microarray-analysis.
