Region Of Interest Tracking

Region of Interest Tracking is an image-analysis technique that follows a defined area, object, or signal across sequential images or video frames. The method identifies the selected region using features such as position, shape, intensity, or texture, then updates its location through frame-to-frame matching or image registration while accounting for movement and changing boundaries. In immunology and infection research, it can quantify immune-cell migration, pathogen spread, intracellular signaling, or changes in tissue regions during time-lapse microscopy. These measurements help researchers compare cellular behavior, evaluate host-pathogen interactions, and connect dynamic visual patterns with infection progression or immune responses.

We have developed a video-rate tracking microscope system that can record and quantify C. elegans behavior at high resolution and high speeds. We have also developed computational methods to reduce the dimensionality of the worm images to a fundamental set of measurements that completely describe the shape of the worm.
