Germ Cell Transplantation and Testis Tissue Xenografting in Mice

このビデオでは、関連する2つの手法を実演します。1つ目は精原幹細胞の移植、すなわちSSC移植であり、2つ目はこれらの組織の異種移植（Xenograft）です。SSC移植は、レシピエントの精巣内でドナーのSSCから精子形成を誘導するために用いられます。これは、GFPまたはLAC Zなどの識別可能な導入遺伝子マーカーを持つドナーの精巣または培養細胞から、SSCsを含む細胞懸濁液を調製することで行われます。次に、受容側雄の精巣を体外に露出させます。この受容側雄は、精巣内の生殖細胞が除去されるように処理されています。その後、受容側雄のEENT管を単離し、taanと混合したドナー細胞を注入します。青色染料。最終的に、レシピエントの精細管上皮がコロニー形成されます。移植後2～3ヶ月で、ドナー由来の生殖細胞による精細管のコロニー形成が認められます。この例では、レシピエント精巣のlacZ染色により、lacZを発現するドナー細胞のコロニー形成が確認されています。このビデオで紹介する2番目の手法は、精巣組織の異種移植に関するものです。精巣組織ラフティングは、未成熟なドナーからの精巣断片を欠損のあるマウス宿主へ移植することにより、ドナー種からの精巣発達および精子形成を導入するために用いられます。これは、まず、ここに示す例のようにブタなどの大型ドナー種の精巣から、移植に適した組織断片を調製することで行われます。次のステップは、免疫不全の成体レシピエントマウスを去勢し、ドナー精巣の断片をレシピエントマウスの背部皮膚下に移植することです。レシピエントマウスはin vivoインキュベーターとして機能し、ドナー種に応じて6〜7ヶ月以内に精巣異種移植片を完全に発達させます。精原幹細胞移植は、ペンシルベニア大学のRalph Brister氏とその共同研究者によって初めて開発され、1994年に初めて報告されました。その後、本手法は広く応用されるようになり、幹細胞の自己複製を制御するシグナル伝達経路の解明など、生殖系幹細胞生物学における重要な疑問を解決するためのゴールドスタンダードなバイオアッセイとなっています。私たちが異種移植というアイデアを最初に得たのは、非げっ歯類（例えばブタ、霊長類、ヒトなど）のドナーから単離した精原幹細胞をマウスの精巣に移植しても、ドナー由来の完全な精子形成に至らないことに気づいたときでした。単離した生殖細胞ではなく、精巣組織の小さな断片を移植して精巣の微小環境を維持することで、この制限を克服でき、系統的に離れた種のドナーからマウス宿主内で完全なドナー由来の精子形成を誘導できることを明らかにしました。これは2002年に我々によって初めて報告されました。生殖細胞移植プロトコールでは、注入する細胞懸濁液の調製が必要です。調製の詳細については、執筆された論文を参照してください。
