上清を以前に収集した上清とプールし、新しいペレットを50マイクロリットルの分離溶液に再懸濁します。エキソソームを単離するためのSDSページ分析では、まず、プールした上清11ミリリットルを3.3ミリリットルのexo quick TC試薬と摂氏4度で少なくとも12時間インキュベートします。インキュベーション後、サンプルを遠心分離し、上清を新しいチューブに移してSDSページ分析を行い、尿中エキソソームを含むペレットを使用してD-N-A-R-N-Aまたはタンパク質を抽出するか、摂氏マイナス80度で保存します。
NanoDrop分光光度計を使用して、2マイクロリットルのサンプルで260ナノメートルと280ナノメートルの吸光度を測定することにより、各抽出物のDNAとRNAの濃度を決定します。未処理の尿を含むレーンでは、SDSページ分析とウェスタン血液分析を使用したバイオマーカー検出の修正複製が示され、90〜100キロダルトンに対応する最初の尿中ペレットタンパク質の禁止が観察され、最終上清およびエキソソームペレットレーンでそれぞれTAM馬の落下タンパク質の存在と除去が示されました。TAMウマ落下タンパク質バンドは観察されず、初期のステップでTAMウマ落下タンパク質が除去され、エキソソームペレットの収量が増加したことを示しています。
