内皮機能不全は、内皮依存性血管拡張6における障害を特徴とする、血管老化7の顕著な特徴である。年齢と共に、血管拡張を促進する内皮の能力は、主として、生物学的利用能の低下、ならびに血管平滑筋細胞における内皮および sGC におけるエノスの異常な発現および機能に起因して徐々に減少している8,9,10. バイオアベイラビリティを低下させることは、内皮依存性血管収縮剤の産生を増強する。高齢の動脈では、内皮機能不全が培地で過形成を引き起こし、壁厚の顕著な増加によって反射されるように、ヒトにおいて認められる高血圧およびアテローム性動脈硬化において血管肥厚を連想する中間核の数患者13、14。加えて、肥満、糖尿病または高血圧のような病態生理学的条件は、内皮機能障害の発症を加速する15,16.
血管周囲脂肪組織 (PVAT) は、血管構造および機能17を調節するために多数のアディポカインを放出する。PVAT の抗収縮効果は、アディポネクチン、NO、過酸化水素、硫化水素18、19、20などの弛緩因子によって媒介される。しかし、場所および病態生理学的条件に応じて、PVAT はまた、様々な動脈21における収縮反応を増強することができる。PVAT によって産生されるプロ収縮物質には、アンジオテンシン-II、レプチン、レジスチン、および ROS22,23が含まれる。 孤立した血管の研究のほとんどでは、PVAT は、血管系のための単純な構造的なサポートとして考えられているため、血管のリングセグメントの調製中に除去しました。脂肪機能障害は高血圧および関連した心血管合併症24のための独立した危険因子を表すので、血管の応答性を調査するとき、血管を取り巻く PVAT は考慮されるべきである異なる動脈。
