ライブサンゴ細胞のソート用のフローサイトメーターを設定するには、フローサイトメーターソフトウェアで新しいプロジェクトテンプレートを開き、レーザーパネルで分析に適したレーザーを選択します。次に、適切なプロットに側面散布図と前方散布図を作成し、格子および解析戦略を設定します。フローサイトメトリック分析と生きたサンゴ細胞の選別のために、細胞計のサンプルチャンバーにコントロール未染色細胞サンプルをロードし、読み取りプロセスを開始します。
サンゴ細胞は特に壊れやすいので、細胞のリシスを防ぐために細胞メーターの圧力を低く保ちます。散布図では、写真の乗数電圧を調整してポイントを中央に配置し、イベントの記録を開始します。前方散布図では、前方散布図の 10 から 4 番目のマークの周りに、セルの周囲にゲートを作成します。

残骸を洗い流すために適切な時間が経過したら、所望の集団を選別して10,000から数百万個の細胞を収集します。新しい染色、種、またはソーターを使用するたびに、関心のある少なくとも20,000個の細胞を500マイクロリットルの染色媒体に分類して、対象の集団に対して純度チェックを行い、ソートされた細胞を再分析して、最初のソートに使用されるゲート内でイベントが読み取られていることを確認します。インビトロ研究では、並べ替えられた細胞をインキュベーターまたは滅菌環境に移すまで氷の上に保管します。
サンゴ細胞と同じ形状や粒度を共有しない他の非細胞粒子を持ち込むためには、前方および側面散布図でゲート選択を作成することにより、解析から除去することができます。死細胞は、ヒストグラムを用いて染色されていない細胞懸濁液のDAPIシグナルを比較し、高DAPI発現細胞を外に出すことによって除外することができる。並行して、自己蛍光共生症をホストする細胞は、遠赤色チャネル内の高いシグナルを有する細胞に対して行うことによって除去することができる。
この反復ゲーティングプロセスの後、残りの細胞は共生性を欠く無傷の生きたサンゴ細胞集団を表す。これらの細胞は、活性酸素種活性および/またはリソソーム含有量などの特徴を染色することによって、異なる細胞亜集団に分類することができる。一部のセルは他のセルよりも脆弱であり、このプロセスを可能な限り慎重に実行することが重要です。
