腸粘膜は、単層の柱状腸上皮細胞(IEC)、下層の層の層の下層の下層のプロプリア免疫細胞および筋性粘膜を包含する。栄養素の吸収におけるその役割に加えて、腸上皮は、発光コメンサル細菌、病原体、および食事抗原から身体内部を保護する物理的な障壁である。また、IECとラミネラプロプリア免疫細胞は、コンテキストおよび刺激に応じて耐性または応答のいずれかを誘導する免疫応答を調整します。上皮バリアの破壊は、病的粘膜炎症の発症に先行し、潰瘍性大腸炎およびクローン病1、2、3、4、5、6、7の両方を包含する炎症性腸疾患(IBD)に寄与することが報告されている。潰瘍性大腸炎を有する個体は、多形核好中球(PMN)の過剰な経上皮移行(TEpM)を形成し、陰窩膿瘍を形成し、疾患8、9の重症度に関連していた知見である。侵害された上皮バリア機能と過剰な免疫応答はIBDの特徴であるが、腸粘膜への腸透過性および免疫細胞の配置の定量的評価を行う実験的インビボアッセイが不足している。
腸上皮透過性およびPMN TEpMを研究するために用いられる最も一般的な方法は、半透過性多孔膜挿入物10、11、12に培養されたIEC単層を用いたex vivoチャンバーベースのアプローチを採用する。上皮バリア完全性は、エピテリア電気抵抗(TEER)またはフルオレセインイソチオシアネート(FITC)標識デキストランの経上皮電気抵抗(TEER)の測定値のいずれかによって監視される。同様に、PMN TEpMは、典型的には、下のチャンバ16に添加される化学誘引剤に応答して研究される。PMNは上部チャンバに置かれ、インキュベーション期間の後、基底コンパートメントに移行したPMNが収集され、定量化される。これらの方法は便利で、実行しやすく、非常に再現性がありますが、明らかに還元的なアプローチであり、必ずしもin vivo条件の正確な反映を表すものではありません。
マウスにおいて、腸内細胞透過性を研究する一般的なアッセイは、FITC-デキストランの経口ギャバジおよびその後の血清中のFITC-デキストランの外観の測定による、13、17である。このアッセイの欠点は、地域の腸管の寄与ではなく、消化管の全体的なバリア完全性の評価を表していることである。また、エバンブルーは、生体内18における血管漏れを評価するために一般的に使用され、また、マウスおよびラット19、20、21における腸粘膜透過性を評価するために採用されている。腸粘膜におけるエバンスブルーの定量化は、一晩ホルムアミドでインキュベーションを採用する組織からの抽出を必要とする。したがって、同じ組織を使用して腸内皮透過性および好中球浸潤を研究することはできない。
ここでは、生体内での大腸粘膜透過性および白血球経上皮移動に関する再現性データを収集するために必要な動物の数を減らす簡単なプロトコルを強調する。したがって、さらなる分析のために収穫できる腸ループの完全性を損なうことなく、血清中で容易に検出できるFITC-dextransの使用をお勧めします。なお、腸連結ループは、細菌感染(サルモネラ菌、リステリア単球遺伝子および大腸菌など)を研究するために様々な種(マウス、ラット、ウサギ、子牛を含む)で使用されてきた22、23、24、25、腸透過性26;しかし、我々の知る限りでは、IBDに一般的に関与する腸内の特定の領域(イリウムや結腸など)におけるPMN TEpMのメカニズムを調査する研究は行っていません。
ここでは、イラミウムまたは近位結腸の良好な血管化および外形化された腸セグメントを採用した堅牢で信頼性の高いマイクロサージカルin vivo法であるマウス腸管ループ(iLoop)モデルについて説明する。iLoopモデルは生理学的に関連しており、麻酔下で生きているマウスの腸バリア完全性およびPMN TEpMの評価を可能にする。2つの用途を実証する:1)iLoop2におけるイントラミン投与後の4kDa FITC-dextranの血清レベルの定量化)強力な基調剤ロイコトリエンB4(LTB4)27の内環注入後のiLoopルーメン中のトランスマイグレーションPMNの定量化。さらに、ILoopモデルを利用して、JAM-a-nullマウスまたはIEC上でJAM-Aの選択的損失を有するマウス(Villin-cre;Jam-a fl/fl)は、コントロールマウスと比較して、腸透過性および好中球の転化15、28、29、30、31に対する緊密な接合関連タンパク質JAM-Aに対する主要な寄与を報告した以前の研究を裏付けることができる。
iLoopモデルは、体外アッセイを裏付ける高機能・生理学的方法です。さらに、これは、ケモカイン、サイトカイン、細菌病原体、毒素、抗体および治療薬を含むループ内腔に注入することができる様々な試薬の研究を可能にする多目的な実験モデルである。

Functional Assessment of Intestinal Permeability and Neutrophil Transepithelial Migration in Mice using a Standardized Intestinal Loop Model

腸粘膜は、単一の上皮細胞層が並び、光細菌や外因性物質の通過を防ぎながら、栄養素や水の超細胞輸送を可能にする動的バリアを形成する。この層の違反は、発光内容物への透過性の増加と免疫細胞の募集をもたらし、どちらも炎症性腸疾患(IBD)を含む腸内の病理学的状態の特徴である。 
多形核好中球(PMN)の上皮バリア機能と経上皮移行(TEpM)を調節するメカニズムは、定量的分析を可能にするインビボ法の実験不足のために不完全に理解されている。ここでは、腸管または近位結腸の外形化された腸管セグメントを採用した堅牢なマウス実験モデルについて説明する。外装された腸ループ(iLoop)は完全に血管化され、上皮細胞単層間の透過性およびPMNの移動を研究するために一般的に使用されるex vivoチャンバーベースのアプローチよりも生理学的な利点を提供する。
