このプロトコルでは、AAV2ベクターは、バキュロ ウイルス (BV)-AAV2-緑色蛍光タンパク質(GFP)または治療遺伝子およびBV-AAV2-Rep-cap感染したSf9細胞を懸濁培養中にSf9細胞と共培養することによって産生される。AAV粒子は、洗浄剤を使用して細胞から放出され、精製され、親和性カラムクロマトグラフィーによって精製され、接線流ろ過によって濃縮される。
