1. トレンチサンプリングによる現場掘削
<ol>
	<li>商業用サトウキビ雑種(以下「品種」と呼びます)を、120 cmの列間隔で植えられた2つの芽のセットから畑で育て、90 cmの列に植えます。これは、推奨される実践パッケージ(POP)に従って、良好な作物の確立と成長を確保します。</li>
	<li>作物の成熟期の終わりに、掘削機を使用して畑に溝(深さ1.5m、幅1.0m)を掘ります。連続的なウォータージェットを通じて、根に損傷を与えることなく根域からの土壌がきれいになることを確認します(図1)。</li>
	<li>付着した土壌が緩んだら、根系と一緒にサトウキビを根こそぎにし、実験室に持って行き、根の数、根の長さ、体積、重量を手動で測定します。</li>

2.サンプリングエラーを低減するルートコアサンプラー
<ol>
	<li>サトウキビの根のサンプリングには、高さ61cm、直径16cm、重さ8kgの円筒形の根芯サンプラーを使用し、軟鋼(MS)材料を使用して現場でのサンプリングを行います。サンプラーの下端に鋭いエッジを設けて、土壌に挿入しながら簡単に浸透できるようにします。サンプラーを持ち上げるために、上部に直径3cmのカラーを用意します(図2)。</li>
	<li>サトウキビの品種は、120 cmの列間隔で植えられ、列内に90 cmの2つの芽のセトから畑で育てられ、推奨されるPOPに従って、良好な作物の確立と成長を確保します。</li>
	<li>作物の成熟段階で、サンプラーの上端を一次シュート/サトウキビに固定し、希望の土壌深さ(45 cm)に達するまで連続してハンマーで打ちます。全体の土壌塊をサンプラーに持ち上げ、流水で慎重に洗浄して付着した根を分離します。</li>
	<li>根を徹底的に洗浄した後、手動測定で体積、表面積、長さ、重量を記録するとともに、透明なトレイに根を広げて、参照されたソフトウェアを使用して対応するデジタル化された画像をスキャンして分析します( 資料の表を参照)。</li>

3. 異なる表現期でのサンプリングを容易にするルート表現型構造
<ol>
	<li>サトウキビの根をサンプリングするための寸法4.5 m x 10.1 mの3つの隣接するコンパートメントを含む根表現型構造を構築し、側壁を手動で解体して地下の根系(深さ80〜100 cmまで)を明らかにするための準備をします(図3)。側壁は、長さ1.8 m、幅30 cm、厚さ4 cmのプレキャストスラブで構築します。</li>
	<li>構造物を畑の土で埋めて圧縮し、~20 cmのヘッドスペースを残し、土壌の通気を容易にするための適切な排水穴を設けます。</li>
	<li>Saccharum officinarum L.、Saccharum spontaneum L.、Saccharum barberi Jesw.、Saccharum sinense Roxb.、Saccharum robustum Brandes、Jeswiet ex Grasslを含む生殖細胞クローンの芽チップを播種します。(以下、「Saccharum spp.クローン」と呼びます)そして、発根培地(赤土:農場の庭の肥料:砂= 2:2:1)を含むプロトレイで30日間発芽させます。良好な作物の確立と成長を確保するために推奨されるPOPに従って、90 cmの列間隔で、列内に60 cmの列間隔で、均一で健康な沈殿物を構造に移植します。</li>
	<li>形成期(植え付け後60〜120日[DAP])および大成長期(120〜150 DAP)の間に、プレキャストスラブで作られた側壁を手動で取り外し、続いてウォータージェットを連続的に噴霧して根を露出させます。</li>
	<li>根系全体を根こそぎにして実験室に持ち込み、長さ、体積、重量を手動で測定し、根を透明なトレイに広げて、参照されたソフトウェアで対応するデジタル化された画像をスキャンして分析します( 材料の表を参照)。</li>
	<li>コンパートメントの 1 つで灌漑を差し控えて干ばつストレスを課し、2 番目のコンパートメントの排水穴を塞いで土壌の飽和を維持し、浸水ストレスをシミュレートします。推奨されるPOPに従って3番目のコンパートメントを灌漑し、フィールド容量を維持し、制御として機能します。</li>

4. 根圏生物学を研究するための植物の水耕栽培
<ol>
	<li>サトウキビの栽培に適した環境制御された温室で、自社製の水耕栽培システムを製作し、根の生物学の細部を研究します。~15 Lの改質ホーグランド養液(表1)を、水槽用ポンプ(図4)による曝気を行いながら、20 cm x 20 cm x 50 cmのガラスタンクに加えます。</li>
	<li>サトウキビ品種と Saccharum spp.クローンのつぼみチップを播種し、堆肥化されたコイアピスを含むプロトレイで30日間発芽させます。均一で健康な沈殿物を水耕栽培タンクに移植し、タンクごとに3回の沈殿の頻度で、根全体を栄養溶液に配置するように注意します。根が光にさらされないように、タンクを黒い布で覆います。ガラスタンクの縁にプラスチックメッシュ(20 cm x 20 cm)を使用して、植物を直立させます。</li>
	<li>発芽期(60日)の終わりに、光合成活動のピーク時に、無傷の植物の根を50 mLのトラップ溶液(滅菌二重蒸留水)に4時間(0800時間から1200時間)浸して、根の滲出液を収集します。収集した溶液をWhatman濾紙でろ過し、次に陰イオン交換で満たされたガラスカラムに通し、続いて陽イオン交換樹脂12を充填します。溶出した画分を蒸発させて乾燥させ、さらなる処理まで-20°Cで保存します。</li>
	<li>処理した根滲出液サンプルをHPLCで分析して有機酸12 を測定し、分光光度法で総フェノール類13、タンパク質14、糖15、およびアミノ酸16 を標準プロトコルに従って推定します。</li>
	<li>週間隔で根の成長を監視して、根の先端の色素沈着と根毛の密度を記録します。酵素、ペルオキシダーゼ17 およびスーパーオキシドジスムターゼ18、および総フェノール含有量19 の活性を、標準プロトコルに従って 3 か月目に評価します。</li>
	<li>滅菌外科用ブレードを使用して、一次根から根の先端までの縦方向のスライスを与えることにより、根の損傷に対する反応を評価し、変化を定期的に監視します。</li>

サトウキビ品種の根の形態学的形質の多様性 トレンチサンプリングによってフィールドから発掘され、水耕栽培のセットアップで成長したCo 62175の根系の代表的な画像を、図5A、Bに示します。Co 62175 と Co 99006 の品種では長い根 (~100 cm) が観察され、Co 99006 は最も高い根の重さ (127 g・clump-1) を記録しました。根形質は根芯サンプラーを使用して記録され、Co 62175 は累積根長、根表面積、および根の量に関して Co 99006 と比較して優れた根系を持っていることが明らかになりました (図 5C-E)。ただし、平均根の直径は、Co 99006 の方が Co 62175 よりも高かった (図 5F)。
