まず、間葉系幹細胞を90%以上のコンフルエントまで培養します。各ディッシュの培養液を8 mLのα-MEMを添加したものと交換します。48時間の培養後、培養液を50 mLの遠沈管に完全に集めるために、細胞を均一に増殖させる必要があります。

次に、トラッキングアナライザーと互換性のあるソフトウェアを起動します。サンプルセルに蒸留水を満たし、装置がセルチェックを開始できるようにします。準備した標準溶液で機器を校正します。
ソフトウェア検出インターフェイスに表示されるパーティクル数が 50 〜 400 の間、できれば 200 前後であることを確認し、[OK] をクリックします。次に、サンプルセルを5mLの蒸留水ですすいでください。サンプル分析の前に、1 mLの蒸留水でチャネルを洗い流してください。
ソフトウェア検出インターフェイスに表示されるパーティクル数が 10 未満であることを確認します。1.5 mLのコニカルチューブで、細胞外小胞を250 uLのPBSで再懸濁します。別の1.5 mLコニカルチューブで、PKH26色素の作業溶液を調製します。
再懸濁した細胞外小胞を作業溶液と混合します。室温で5分間静置してから、EVが枯渇したFBSを500 uL加えて反応を停止します。マイクロベシクルの場合は、摂氏4度で30分間16, 000 x gで遠心分離します。
上澄み液を捨てる。1 mLのPBSを加えて残留物を洗い流します。再度遠心分離し、上清を捨て、未結合色素を除去する。
マイクロベシクルの場合は、摂氏4度で30分間16, 000 x gで遠心分離します。残留物を200 uLのPBSで再懸濁します。20 uLの懸濁液をスライドに滴下し、蛍光顕微鏡で観察します。
細胞外小胞を200 uLのPBSに再懸濁し、ヘパリン溶液と10:1の容量比で混合します。マウスを尾静脈イメージングシステムに置きます。スイッチを押してレバーを持ち上げます。
尾静脈イラストレーターの助けを借りて、尾静脈を通して細胞外小胞懸濁液の体系的な注射を行います。NTAを用いた粒度分布の解析により、ヒト間葉系幹細胞由来のエクソソームのサイズは40〜335 nmの範囲であり、ピークサイズは約100 nmであることが示されました。また、マイクロベシクルのサイズは50〜445 nmの範囲であり、ピークサイズは150 nmでした。
エキソソームの形態学的特徴は、それらが典型的なカップ形状を示すことを示した。細胞外小胞はPKH26によって効率的に標識され、これは標識ペレットとして肉眼視によって観察され、蛍光顕微鏡によって観察された。間葉系幹細胞の不明瞭な}に保持された放出されたEVを完全に収集するには、細胞を均等に吹き飛ばす必要があるため、上清の収集中はオペレーターに注意を払う必要があります。
