細胞密度がミリリットルあたり10〜6セルの2倍、または10〜6セルの2.5倍を超える場合は、より大きな容器に移し、ミリリットルあたり5倍の10〜5セルに希釈します。上下にピペッティングして各ウェルの内容物を穏やかに混合します。マイクロバブルを含むウェルの内容物全体を取り除き、1.5ミリリットルのチューブに移します。
400マイクロリットルのカルシウムフリーおよびマグネシウムフリーのDPBSで各ウェルを洗浄し、溶液を1.5ミリリットルのチューブに移します。次に、チューブを400gで室温で5分間遠心分離します。上清を吸引し、細胞ペレットを50マイクロリットルの分離バッファーに再懸濁した。
次に、活性化と枯渇のために50マイクロリットルの細胞懸濁液をそれぞれ25マイクロリットルに分割します。活性化および枯渇抗体染色カクテルで細胞を染色し、暗所で室温で10分間インキュベートします。1ミリリットルの分離バッファーを加え、穏やかに混合し、遠心分離機で余分な抗体を洗い流します。
上清を完全に吸引します。細胞ペレットを1ミリリットルの分離バッファーに再懸濁し、フローサイトメトリー分析用の適切な容器に移します。生存可能なT細胞数および導入遺伝子陽性T細胞の増加は、対照試料とマイクロバブル共刺激を受けた細胞との間に観察された。
エフェクター細胞集団の増加もマイクロバブルサンプルで観察されました。生存可能な活性化T細胞は、増加した早期活性化マーカーCD69、および中期から後期活性化マーカーCD25を発現する。枯渇マーカーPD1陽性細胞の総数と割合も、マイクロバブル共刺激を受けた細胞サンプルで有意に高かった。
