Sequential Blood Collection from Inferior Vena Cava Followed by Portal Vein to Evaluate Gut Microbial Metabolites in Mice

上記の方法を用いることで、IVCとPVから血液サンプルを同じマウスで順次採取することができ、成功率は95%を超えています。採取される血液サンプルの平均量は、IVCで0.25 mL、PVで0.35 mLです。
ガスクロマトグラフィー質量分析法(GC-MS)を用いて、糞便、PV血液、IVC血液中のSCFAsの濃度を測定し、腸内細菌の発酵によって生成される大腸コンパートメントからアセテート(C2)、プロピオン酸(C3)、酪酸(C4)の吸収と通過を追跡できました。
具体的には、これらの実験では、マウスのSCFAs産生に及ぼす食物繊維の摂取の影響を調査しました。マウスに高繊維食(食物繊維35%)を1週間または2週間与え、食物繊維を含まない食餌を与えられたマウスと比較しました。高繊維グループでは、繊維発酵細菌分類群の増加が予想され、観察されました(データは示されていません)。次に、結腸内容物、PV血、および全身循環(IVC)血液のSCFAsレベルを調べました(ミシガン大学のメタボロミクスコアファシリティのGC-MSで定量化)3 (図2)。PVサンプルでは、食物繊維への曝露によるSCFAの一貫した増加を検出することができましたが、結腸糞便サンプルのSCFA測定値は、PVおよびIVC血液測定値と比較して、マウス間ではるかに変動が大きかった( 図2の酢酸、プロピオン酸、および酪酸の結腸糞便とPV/IVC血液)。この変動性は、糞便中のSCFAの非均質な分布によるものか、または腸上皮によるSCFAの変動吸収を反映している可能性があります。いずれにせよ、宿主の腸外生理機能に対するSCFAの影響を調査する目的では、PV内および末梢循環のレベルが研究により関連性が高い可能性があります。
PV内のSCFAレベルを全身IVC血中のレベルと比較することにより、肝臓による酢酸とプロピオン酸の吸収レベルも観察することができました(図2A、B)。肝臓を通過した後に全身血中に残るアセテートの割合(PVと比較して)は、プロピオン酸の割合よりも高くなります(PVアセテートの50%以上が末梢循環に入ったのに対し、プロピオン酸は30%未満)。これは、プロピオン酸が肝臓に吸収または使用され、体循環に入るレベルが低いのに対し、酢酸は筋肉組織13,20などの他の末梢組織で吸収または使用される可能性があるという以前の報告と一致しています13,20(図2A、B)。それにもかかわらず、いくつかのグループからの以前の研究は、体循環に吸収されるプロピオン酸のレベルが、遠隔臓器の生理学および病理学に有意に影響を与えることができることを示している21,22,23,24。
さらに、PVと全身血液の両方の酪酸濃度は、酪酸とプロピオン酸の糞便濃度は非常に類似しているにもかかわらず、プロピオン酸よりもはるかに低いことがわかりました。これらの発見は、腸で生成された酪酸が結腸で主に吸収され、腸上皮によって局所的に使用されるという以前の研究を確認しました。図11(図2C)。 
通常、腹部解剖中に、PVの寸法がIVCよりも小さいことが観察されます(図3A)。しかし、PVの周りの縫合糸を結束し(肝椎骨より約4mm下)、血管を結紮すると、PVのサイズが大きくなり、PV結紮前のIVCの元のサイズとほぼ同じ直径になります (図3B)。図からわかるように、IVCはPVライゲーション後にわずかに崩壊します(図3B)。全体として、このアプローチは、マウスのPVからの血液の最大限の収集を容易にします。私たちの経験では、最初にPV血液を採取し、次にIVC血液を採取すると、結果よりも最適ではありませんでした。これは、PV採血後にIVCの著しい崩壊が観察されたためです。
