Measuring Carbon Content in Airway Macrophages Exposed to Carbon-Containing Particulate Matters

固定液で保存および処理された喀痰は、形態学的検査中に光学顕微鏡下で無傷のマクロファージ形態を示しました。マクロファージは、透明、円形、または腎臓の形をした、染色しやすい細胞核を示しました。染色後、細胞核は青紫色に見え、細胞質は淡いピンク色または水色でした。顕微鏡的な磁場は最小限の不純物を示し、細胞の同定を容易にしました。細胞内では、黒色炭素粒子が小さな凝集体ではっきりと見え、炭素粒子の定量分析を可能にしました。
喀痰マクロファージの代表的な画像を図1に示し、さまざまな細胞内のさまざまな数の炭素粒子を示しています。たとえば、図1Aは炭素粒子を示していませんが、図1Dは最も高い濃度を示しています。CCAMの値は、マクロファージ(A-D)でそれぞれ0.00%、2.24%、6.64%、34.61%と測定されました。CCAMパラメータ決定プロセスでは、マクロファージサイズの変動や炭素粒子と細胞核との重なりから生じるバイアスを軽減するために、各マクロファージの炭素含有量の定量的評価が行われました。CCAMの計算と解析は、図2に示すように、研究対象者あたり50のマクロファージに対して行われました。
この研究では、特定のアセチレンカーボンブラック工場のカーボンブラックパッカー(CBP)を曝露群として選択し、有害な要因への明らかな職業的曝露がなかった地元の水プラントのウォーターポンプ労働者を対照群(非CBP)として募集しました。 図 2 に示す結果は、対照群と比較して、カーボンブラック包装労働者の CCAM が高いことを示しています。
