薬用植物同定のためのdnaバーコードの応用

Application of DNA Barcoding to Identify Medicinal Plants

サンプルDNAの品質
OD260/OD280の吸光度比の範囲は1.80〜1.84であった。分光光度計で測定した各サンプルのDNA量は100 ng/μLを超えており(表3)、サンプルDNA抽出の品質が良好であることを示しています。これは、抽出プロセス中にサンプルがタンパク質、RNA、または試薬によって汚染されておらず、サンプルの品質が良好で、ダウンストリームのPCR増幅に適していることを示しています。
PCR解析
ゲル電気泳動は、3つの並行実験サンプルのPCR産物に対して行った。セットアップは、 図3に示すように、マーカー、サンプル1〜3(AS-1、AS-2、AS-3)、およびネガティブコントロール(0)でした。AS-1、AS-2、AS-3のサイズはいずれも500 bpで、1つの明るく明確なバンドがあり、0にはバンドはなく、抽出されたDNAがITS2プライマーを使用して500 bpに増幅されたことを示しています。
シーケンシング結果のデータベース比較
シークエンシングの結果は、GPGDデータベースの種同定結果と同じNCBIデータベースのBLAST同定結果を組み合わせることで、100%の配列類似性を持つ A. sinensis を同定しました(表4)。BLAST の結果は、最も高いパーセント ID スコアと最も低い e 値を使用して決定されました。種の同定は、最も高い最大同定率に単一の種が含まれ、&gt;99%のスコアを獲得した場合に成功したと見なされました。スコアが99%≤場合は、シーケンシングファイルの品質、またはPCR産物が汚染されていないかを確認してください。
系統樹に基づく種同定の解析
その結果、構築されたニュージャージー州の系統樹では、4種すべてが別々のクレードに集まっており、アウトグループとしての P.praeruptorum は Angelica クレードから分離されていることが示されました。スプライシング配列は A. sinensis OR879715.1 クレードでクラスター化され、クレード支持値 (ブートストラップ) は 100 であり、他の 2 種、 A. dahurica と A. cartilaginomarginata はそれぞれ別々のクレードにクラスター化され、スプライシング配列が A. sinensis であることが証明されました (図 4)。
