A Mouse Model of Mechanotransduction-driven, Human-like Hypertrophic Scarring

HTSプロトコルの効果的な使用を明確に示し、成功した「肯定的な」結果を特定するために、図3Aに示すようにモデルが確立されました。代表的な研究では、図3B、Cに示すように、図3B、Cに示すように、切開部全体に人間のようなレベルの機械的ひずみが誘発されるNo Stretch Control(n = 6)とMechanical Stretch HTSグループ(n = 6)の2つのグループがありました。図3A-Cに示されている実験計画内で、POD5、9、15、および19の傷跡のグロス写真を撮影しました。瘢痕をImageJで追跡し、異なる時点における瘢痕の平均幅を分析しました(図3D)。平均瘢痕幅は、図3Eの時間およびグループ間でグラフ化されました。POD 5 で機械的ひずみを適用した後、Stretch Group は Stretch と Control の間で傷跡に大きな差がなかったため、Stretch Group は Control よりも有意に広い瘢痕を示し、POD 9 でピーク幅は 0.99 mm に達しました (p = 0.045)。この有意に大きい瘢痕幅は、対照群と比較して、POD 15 (p = 0.043) および 19 (p = 0.045) で継続しました。グロス撮影は、HTSデバイスを取り外す必要がある日(つまり、HTSデバイスが最大拡張に達し、再度取り付ける必要がある日、または摘出中に)にのみ撮影することをお勧めします。このプロトコルでは、複数の時点にわたる画像と瘢痕幅の分析により、モデルの創傷の進行を読者がより明確に把握できます。
