ゼブラフィッシュの胚および幼生における高分解能細胞移植

High-resolution Cell Transplantation in Embryonic and Larval Zebrafish

移植実験の結果は、移植後の適切な時点における宿主動物の蛍光標識ドナー細胞を蛍光顕微鏡で可視化することで直接観察することができます。ここでは、個々の前迷走神経ニューロンを3dpfで移植しました。その後、宿主動物を12時間または48時間インキュベートし、麻酔をかけ、ガラスカバースリップ上にLMAにマウントし、共焦点顕微鏡でイメージングしました(図5)。移植後12時間(hpt)で、移植に成功したドナーニューロンを観察します(図5A)。細胞は宿主迷走神経運動核の前部にある宿主迷走神経ニューロンの中に位置しているため、細胞の正しい位置を確認できます。また、細胞は無傷で健康に見えるが、これはいくつかの短い神経突起の延長によって示される(図5A)。移植後2日(dpt)で、単一のニューロンが宿主核の正しい位置に正常に移植されたことを再び確認できます(図5B)。この時点で、ニューロンが新しい軸索を第4咽頭弓まで伸ばしたことが観察されます(図5B)。直接観察すると、手順が失敗したことも明らかになります。この例では、ドナーニューロンが存在しない2dptの宿主を観察します。むしろ、移植後に死んだドナーニューロンの断片である可能性が高い小さな緑色の斑点が明らかです(図5C)。移植の成功率は、ユーザーの経験と移植された細胞の数と種類によって異なりますが、3-4 dpf33で行われた迷走神経運動ニューロン移植の成功率(2-3 dptで宿主の軸索を伸ばした生存ニューロンの存在として定義される)は66%(n = 453回の移植)を経験します。
