Unveiling Histone Proteoforms using 2D-TAU Gel Electrophoresis

図1Aでは、ヒストンタンパク質が1Dゲル内でどのように移動するかを観察することができます。これは、最初の次元の実行時間を理解するための重要な準備ステップです。分離の適切な時間が決まったら、2 番目のディメンションを実行します。ヒストンプロテオフォームの2Dマップは、独特のトポグラフィーパターンを示しています(図1B)。図1Bの各スポットは、特定のヒストンタイプを表しています。各ヒストンプロテオフォームが見つかるゲルの正確な領域を観察します。各ヒストンプロテオフォームの質量分析同定は、Perri et al.32で報告されています。したがって、この状態の各スポットは、特定のヒストンタイプに関連付けられています。標準的なスポットと並んで追加のスポットが存在することは、新しい変更の存在を示している可能性があります。ゲル画像はデンシトメトリーを使用して取得でき、得られた画像は画像解析ソフトウェアを使用して分析できます。
2D TAU マップの信頼性は、スポットの丸みに依存します。ランが正しく実行されている場合、スポットは個別のシグナルとして表示され、水平および垂直のストリーキングは見えません。逆に、サンプル、溶液、およびアガロースのオーバーレイ中の不純物は、ゲルに縞や縞模様を引き起こす可能性があります。ゲルの品質は、画像のデジタル化とそれに続く定量的および定性的な分析に影響を与えます。さらに、ウェスタンブロット解析やMS調査の成功にも影響を与える可能性があります。この分析法の信頼性を検証するために、分離されたヒストンプロテオフォームのニトロセルロースメンブを、一次抗体アセチル化リジン(Ac-K2-100)ウサギmAb32 でプローブしました(図2)。抗ウサギHRP二次抗体を使用して、ヒストンシグナルを検出しました。予想通り、デンシトメーターを使用すると、ブロット中に存在する各ヒストンバリアントの特異な翻訳後修飾を定量化することができます。
