Time-Lapse Epifluorescence Microscopy Imaging of Pseudomonas aeruginosa and Staphylococcus aureus Heterogeneous Phenotypes

レポータープラスミドの 緑膿菌 および 黄色ブドウ球菌 への導入の成功は、正しい選択的抗生物質の増殖によって示され、コロニーPCRおよび/またはシーケンシングによって確認することができます。改変された株は、目的の遺伝子が誘導されることが知られている条件にそれらをさらすことにより、表現型レポーターとして検証する必要があり、結果として生じる蛍光は、フローサイトメトリー、分光光度法、または落射蛍光顕微鏡法で測定できます(図1)。
その後の実験に使用する抗生物質の用量の選択を容易にするために、目的の 緑膿菌 または 黄色ブドウ球菌 株の濃度依存性抗生物質持続性アッセイを実施します。濃度依存性アッセイでは、通常、抗生物質濃度が低いと初期傾斜が急になり、高濃度ではプラトーまたはそれほど急でない傾斜を持つ二相性曲線になります。ただし、一部の抗生物質と生物種のペアでは、明確な二相性曲線が生じない場合があります。例えば、 黄色ブドウ球菌(S. aureus delafloxacin)の曲線は明らかに二相性であるが(図3A)、 緑膿菌の レボフロキサシン曲線はそうではない(図3B)15。このシナリオでは、MICの少なくとも10倍の濃度を選択します(たとえば、5μg/mL、 これは緑膿菌のMICの約15倍です)15。ただし、15x レボフロキサシン MIC では 緑膿菌 の生存者は ~0.001% しか得られないため、抗生物質を含まないアガロース パッドで細胞が回復する細胞をイメージングする際に持続性を確認したい場合は、1 μg/mL のレボフロキサシン治療を使用します (補足ビデオ 6)。そうしないと、複数の永続性をイメージングするために必要な視野の数が非常に多くなります。
イメージングの開始時には、理想的なサンプルとアガロースパッドの調製は、視野全体にわたって平面的であり、大きな破片、しわ、気泡がなく、単一細胞が均一に分布している必要があります。十分に分布した単一細胞を得るには、サンプルの希釈または再懸濁の最適化が必要になる場合があります。黄色ブドウ球菌の場合、細胞は小さなクラスターを形成する傾向があり、アガロースパッドに播種する前に十分にボルテックスする必要があります(図4および図5)。緑膿菌の場合、細胞は懸濁液中の粘着性のある細胞外マトリックスに包まれた凝集体を形成することがあります。これらのサンプルを徹底的にピペットで動かし、単一細胞をイメージングするために凝集体を破壊する必要があります。
