In Situ Isolation and Culturing of Recalcitrant Soil Bacteria using an Isolation Chip (iChip)

このプロトコルで使用されるiChipは、 図1に示すように設計、3Dプリント、および組み立てられました。説明されているプロトコルの概要を 図2に示します。実験に使用された土壌サンプルは、インドのラジャスタン州にある IIT ジョードプル キャンパスから収集されました。その場所のユニークな 3 語の住所は「communicate.supervising.recurrence」でした。選ばれた場所は、サンプリング当日の気温が40°Cの半乾燥地帯でした。土壌サンプルは真夏に採取されました。サンプルは、表面下温度35°Cで深さ6〜7インチで無菌的に無菌で収集され、収集後24時間以内に処理されるラボに運ばれました。次に、それらを連続的に希釈し、トリプシン大豆寒天培地にプレーティングして、適切な希釈を推定しました。土壌の培養可能な微生物密度は、土壌の 8 ×10 6 CFU でした。土壌懸濁液の10-3 希釈液には、播種用に選択された~8 x 102 CFU / m...
