Comprehensive Analysis of Drug Response using the FLICK Assay

このプロトコルを使用して、HDAC阻害剤Belinostatに対するU2OS細胞の感受性を調査しました。これらの実験は、2 μM SYTOX Greenを使用して死細胞を標識するために行いました(図1A)。キネティック読み取りは、130ゲイン設定の蛍光プレートリーダーを使用して行われました(図1B)。アッセイの終了時に、PBS中の1.5% Triton-X溶液に細胞を2時間溶解しました(図1C-D)。
