本研究は、化粧品、医薬品、および局所的な生物医学的応用のために、cerrado原産の植物から持続可能なマグネシウムナノ粒子を開発することに焦点を当てています。このプロトコルは、持続可能なナノフィトコスメティックの開発、植物ベースのナノ粒子合成、およびスケールアップ可能な化粧品処方の研究に応用可能です。まず、採取したばかりの樹皮を流水ですすぎ、土、ほこり、および表面の不純物を除去します。樹皮サンプルを36 °Cのオーブンで72時間乾燥させます。ナイフミルを用いて、乾燥した樹皮を粉砕して微粉末にします。粉末状の樹皮200 gを三角フラスコに移します。98% ethanolを1000 mL加えます。フラスコを密閉し、遮光条件下で室温にて72時間保存し、その間24時間ごとに緩やかに振とうします。抽出液をまず金属メッシュでろ過し、次いで漏斗を用いて綿でろ過し、残留した不純物を除去します。抗酸化活性を評価するため、100 millimolarのDPPHメタノール溶液を調製します。エタノール樹皮抽出物をメタノールで希釈し、容量比0.1%から5%の範囲の濃度系列を調製します。同様の濃度となるビタミンC溶液を用いて、陽性対照を調製します。96ウェルプレートの各ウェルに、DPPH溶液を100 microlitersずつ分注します。各ウェルに、抽出物またはビタミンC溶液を50 microliters加えます。プレートを室温、遮光条件下で30分間インキュベートします。分光光度計を用いて515 nanometersにおける吸光度を測定し、ラジカル消去活性を算出します。正常ヒト皮膚線維芽細胞を、10%胎児牛血清および1%ペニシリン-ストレプトマイシンを添加したRPMI 1640培地で培養する。細胞を96ウェルプレートに1ウェルあたり3 × 10⁴ cellsで播種し、24時間インキュベートする。培地を100 µLの新鮮な培地に交換した後、50 µLの抽出液を添加する。細胞生存率を評価するため、37 °C、5% CO2条件下で24時間インキュベートする。培地を除去した後、各ウェルに10 µLのMTT溶液を添加する。ホルマザンを形成させるため、4時間インキュベートする。結晶を溶解させるため、イソプロパノール中塩酸を含む60 µLの可溶化溶液を添加する。分光光度計を用いて540 nmの吸光度を測定し、細胞生存率を算出する。滅菌済みの反応チューブに9890 µLの蒸留水を加えます。植物抽出液100 µLと1 M塩化マグネシウム10 µLを加え、最終濃度が10 mMとなるようにナノ粒子懸濁液を調製します。溶液を均質化するために十分に混合し、混合物を75 °Cで18時間インキュベートします。混合物を室温まで冷却した後、ナノ粒子懸濁液約750 µLを動的光散乱測定用のセルに移します。セルを装置にセッ
