私たちの研究は、メラノサイトの老化表現型と、それが外因性皮膚老化にどのように寄与するかを調査することに焦点を当てています。確立された老化モデルでは線維芽細胞が使用されており、メラノサイト特有の生物学的特性を捉えきれていません。本プロトコルでは、ヒト初代メラノサイトにtBHP介在性の酸化ストレスを誘導します。まず、必要な試薬を準備します。メラノサイトを融解し、3日間回復させた後、播種日に細胞のコンフルエンシーが80〜90%に達していることを確認してください。フラスコから培地を吸引します。細胞をDPBSで2回洗浄します。細胞を剥離させるため、trypsin EDTA溶液を2 mL加えます。フラスコをインキュベーターに入れ、37 °C、5% carbon dioxideの条件下で5分間置きます。フラスコを軽く叩きます。次に、顕微鏡下で細胞を観察し、完全に剥離していることを確認します。反応を停止させるため、トリプシン阻害剤を1.5倍量加えます。細胞懸濁液を15 mLのコニカルチューブに移します。DPBSを用いて、懸濁液を最終容量10 mLまで希釈します。細胞懸濁液を相対遠心力100×gで5分間遠心分離します。上清を吸引し、細胞ペレットを目的の量まで培地で再懸濁します。tert-Butyl hydroperoxide（tBHP）処理プレート用には、10 centimeterディッシュあたり600,000個の細胞を播種します。コントロールプレート用には、1プレートあたり400,000個の細胞を播種します。tBHPストック溶液を用いて、DPBSまたは培地で10 millimolarの調製液を作成します。1回の処理につき、100 micromolarのtBHPを細胞に加えて1時間処理します。プレートを前後に動かし、tBHPを培地全体に均一に分散させてください。細胞を37°C、5% carbon dioxideのインキュベーター内に正確に1時間置きます。培地を吸引し、tBHP処理を終了させます。37°Cに予熱したDPBSでプレートを2回洗浄します。プレートに適切な量の新鮮な培地を加えます。プレートをインキュベーターに入れ、次の処理まで少なくとも4時間、細胞を回復させます。同日の後ほど、2回目の処理として処理手順を繰り返します。4日目まで治療レジメンを継続し、その後、細胞老化マーカーを評価するために9日目まで培養を維持します。tBHP処理済みの細胞とコントロール細胞の数を数えます。これらを同じ初期密度で再播種します。ただし、tBHP処理済みのプレートから得られた細胞数が600,000個未満の場合は、回収したすべての細胞を再播種してください。培養4日目、9日目、および15日目に細胞数を計測します。9日目に、PBSで細胞を2回洗浄します。2% formaldehydeおよび0.4% glutaraldehydeを用いて細胞を固定します。固定した細胞をPBSで2回洗浄します。pH 6の染色液中で、固定した細胞をインキュベートします。プレートを遮光して覆い、二酸化炭素不在の条件下で37°Cで18時間インキュベートします。細胞をPBSで2回洗浄します。光学顕微鏡を用いて、青色に染色された細胞を撮影し、その数をカウントします。それを同一視野内の全細胞数で除します。結果を陽性細胞の割合として表します。酸化ストレス誘導後、tBHP処理により、対照群と比較してメラノサイトの増殖が有意に抑制されました。対照群が9日目までに3.1 cPDLに達したのに対し、処理群の細胞は1.4 cPDL（累積細胞分裂回数）に留まりました。15日目までに、処理した細胞は1.9 cPDLでプラトーに達しましたが、対照細胞は5.1 cPDLまで増殖しました。tBHPで処理したメラノサイトは、すべての測定時点で対照群と比較して細胞表面積が増加しており、15日目まで肥大が進行しました。これは、老化細胞に見られる肥大化および扁平化した形態と一致しています。tBHP処理したメラノサイトでは、老化関連β-ガラクトシダーゼ（SA-β-ガラクトシダーゼ）活性の上昇が認められ、陽性細胞率は対照群の3%に対し32%でした。SA-β-ガラクトシダーゼ陽性率の上昇は、細胞老化の表現型と一致していました。免疫蛍光分析では、コントロールと比較して、tBHP処理したメラノサイトにおいてgamma-H2AXレベルの上昇が認められました。処理した細胞は、コントロールよりも強いgamma-H2AXシグナルを示しました。リアルタイム定量PCR分析により、細胞老化関連分泌表現型（SASP）因子の発現上昇が示され、特にinterleukin-8で最大の上昇が認められた。一方、interleukin-6およびmatrix metalloproteinase-1では中程度の増加が見られた。ウェスタンブロット分析により、細胞老化の誘導が確認された。tBHP処理したメラノサイトでは、対照群と比較してLamin B1が有意に減少しました。また、tBHP処理したメラノサイトでは、対照群と比較してリン酸化レチノブラストーマタンパク質のレベルが低下しました。tBHP処理により、メラノサイトにおけるp16およびp21の転写発現が増加しました。このモデルを用いることで、老化メラノサイトが色素沈着にどのように影響し、SASP炎症因子を介して周囲の細胞とどのように相互作用するかを研究することが可能になります。本プロトコルにおける課題は、正確なタイミングの管理です。tBHPは、少なくとも4時間の間隔を空けて1日2回投与する必要があります。本プロトコルに従い、老化関連β-ガラクトシダーゼ染色、免疫蛍光染色、RT-qPCR、および確立された老化マーカーのウェスタンブロッティングを用いて、細胞老化を特徴付けることができます。
