Feedback Inhibition

생화학 반응은 세포에서 끊임없이 일어나며, 보통 반응을 촉진하는 효소의 도움으로 시작 물질을 다른 생성물로 변환합니다. 효소가 없다면, 대부분의 화학 반응이 세포에 도움이 되는 데는 너무 오랜 시간이 걸릴 겁니다!
 
 효소가 반응 속도를 조절하는 데 도움이 되기 때문에, 효소의 활동은 세포에서 적절한 양의 시작 물질, 중간 대사물, 그리고 생성물이 유지되도록 조절됩니다. 물질의 과도한 축적이나 고갈은 세포와 유기체의 건강에 치명적인 결과를 초래할 수 있습니다.
 
 되먹임억제(feedback inhibition)라고 불리는 조절 과정에서, 화학 반응의 생성물은 해당 생성물을 생산한 대사 경로(metabolic pathway)의 이전 단계에 작용하는 효소를 억제합니다. 이때 생성물은 효소의 조절부위(regulatory site)와 결합하는데, 이는 기질(substrate)에 결합하는 활성부위(active site)와 구별되며, 생성물과 효소의 결합으로 인해 생성물 자체 생산이 느려지거나 중단됩니다. 생성물이 조절부위와 결합하면 효소에 형태적인 변화(conformational change)를 일으켜 효소가 기질에 결합하여 화학반응을 촉매하는 것을 막습니다.
 
 이렇게 하면 지나치게 많은 생성물이 세포에 축적되는 것을 방지할 수 있습니다. 생성물 수치가 충분히 낮아지면, 해당 생성물은 더 이상 효소를 억제하지 않게되고 화학 반응은 정상적으로 진행될 수 있습니다.

생화학 반응은 세포에서 지속적으로 발생하며, 일반적으로 속도를 높이는 효소의 도움으로 시작 물질을 다른 제품으로 변환합니다.

Cells use enzymes to run biochemical reactions and make the products they need. These reactions are happening constantly, and enzymes speed them up so they happen fast enough to matter. Enzyme activity also has to be regulated. Cells need the right amounts of starting materials, intermediate metabolites, and final products.

If these substances build up too much or drop too low, the cell can be harmed. That is why cells use control systems to keep metabolic levels balanced. One important control process is feedback inhibition. In this process, the final product of a reaction slows an enzyme from an earlier step in the same pathway.

The product binds to a regulatory site on the enzyme. This site is different from the active site, which is where the substrate normally binds. Binding at the regulatory site changes the enzyme’s shape. That conformational change prevents the enzyme from binding the substrate and keeps the reaction from continuing.

Feedback inhibition helps stop too much product from accumulating in the cell. When product levels fall, the product no longer inhibits the enzyme. The pathway can then start again and continue producing the molecule as needed.
