The Central Dogma

 생물학의 중심원리(central dogma)는 DNA에 암호화되어 있는 정보가 전령RNA(messenger RNA, 줄여서 mRNA)로 전달되어 단백질의 합성을 지시한다고 말합니다. mRNA 뉴클레오타이드(nucleotide) 서열을 아미노산으로 해독할 수 있는 일련의 지시를 유전암호(genetic code)라고 합니다. 이 유전암호(genetic code)의 보편성은 과학 연구, 농업, 의학 분야의 발전을 촉진했습니다.
 
 <h4>RNA는 DNA와 단백질 사이의 잃어버린 고리입니다</h4>
 
 1900년대 초, 과학자들은 DNA가 세포 기능에 필요한 모든 정보를 저장하고 단백질이 대부분의 세포 기능을 수행한다는 것을 발견했습니다. 하지만, 유전 정보를 기능 단백질로 바꾸는 기작은 수년 동안 알려지지 않았습니다. 처음에는 단일 유전자가 암호화된 단백질로 직접 변환된다고 믿었습니다. 그러나 진핵세포(eukaryotic cell)에 대한 두 가지 중요한 발견이 이 이론에 도전했습니다. 첫째, 단백질 생산은 핵에서 일어나지 않습니다. 둘째, DNA는 핵 바깥에 존재하지 않습니다. 이러한 발견은 DNA와 단백질 생산을 연결하는 중간 분자를 찾는 계기가 되었습니다. 핵과 세포질(cytoplasm)에서 발견되고 단백질 생성과 관련된 이 중간 분자는 RNA입니다.
 
 전사(transcription) 중 RNA는 DNA를 주형(template)으로 사용하여 핵에서 합성됩니다. 새로 합성된 RNA는 DNA 가닥과 서열이 비슷하지만, DNA의 타이민(thymine; 티민)은 RNA의 유라실(uracil)로 대체됩니다. 진핵생물에선 이 1차 전사물(primary transcript)은 단백질의 비암호화 영역을 제거하고, 5’ 말단에 모자(5' cap)를 씌우고(RNA 캡핑과정; RNA capping; 5' capping), 3’ 말단에 다중 A 꼬리(poly-A tail)를 추가하는(폴리아데닐화; polyadenylation) 추가 처리를 통해 세포질로 운반되는 mRNA를 만듭니다.
 
 <h4>mRNA 서열을 해석하는 규칙은 유전암호를 구성합니다.</h4>
 
 번역(translation)은 세포질의 리보솜(ribosome)에서 발생하며, 여기서 mRNA에 암호화된 정보가 아미노산 사슬로 번역됩니다. 한 개의 아미노산에 대응되는 세 개의 뉴클레오타이드로 구성된 암호를 코돈(codon)이라고 부릅니다. 어떤 코돈이 어떤 아미노산을 지정하는지 설명하는 일련의 규칙이 유전암호를 구성합니다.
 
 <h4>유전암호는 중복됩니다</h4>
 
 단백질은 진핵생물에서 20개의 아미노산으로 만들어집니다. 세포에 있는 4종류의 뉴클레오타이드(즉 아데닌(A), 유라실(U), 구아닌(G), 사이토신(C) 염기를 가진 뉴클레오타이드; mRNA를 설명하고 있기 때문에 타이민(T) 대신 유라실)를 3개로 모으면 가능한 코돈이 64개(43으로 계산)가 나옵니다. 이렇게 아미노산 개수(20개)와 코돈 개수(64개)의 차이는 개별 아미노산이 둘 이상의 코돈에 의해 암호화될 수 있다는 것을 의미합니다. 따라서 유전암호는 중복된다고 부릅니다. 종종 (항상 그렇지는 않지만) 동일한 아미노산을 지정하는 코돈은 뉴클레오타이드 3개 중 세 번째 뉴클레오타이드만 다릅니다. 예를 들어, 코돈 GUU, GUC, GUA, GUG는 모두 아미노산 발린(valine)을 나타냅니다. 한편 AUG는 아미노산 메티오닌(methionine)을 나타내는 유일한 코돈이며, 단백질 합성이 시작되는 코돈이라 개시 코돈(start codon; 시작 코돈)이라고 부릅니다. 코돈 체계의 중복은 돌연변이의 해로운 영향을 최소화합니다 (예: 돌연변이가 코돈의 세 번째 뉴클레오타이드를 바꿔도 여전히 같은 아미노산이 생성될 수 있습니다).
