The DNA Replication Fork

세포에서 DNA 복제는 항상 복제의 기원이라고 하는 DNA의 특정 위치에서 시작됩니다.
복제가 시작되면 효소 DNA 헬리카제(DNA helicase)가 DNA 가닥을 따라 결합하고 이동하면서 DNA를 풀고 분리합니다. 
DNA의 분리된 가닥에 의해 형성된 Y자형 구조는 2색 갈래처럼 보이며 DNA 복제 부위가 됩니다. 따라서 이 구조는 "복제 포크"로 알려져 있습니다.
이러한 분리된 단일 가닥 DNA 분자는 이중 가닥 헤어핀 루프를 형성하거나 다른 가닥으로 되감기 경향이 있습니다. 
이러한 일이 발생하지 않도록 단일 가닥 DNA 결합 단백질(SSB)은 단일 가닥 DNA에 결합하고 되감기를 억제합니다.
이제 DNA의 노출된 단일 가닥은 상보적인 딸 가닥의 합성을 위한 주형으로 작용할 수 있습니다.

E. coli와 같이 작은 원형 DNA를 가진 유기체는 종종 단일 복제 원점을 갖습니다. 따라서 초기 개방 위치에서 멀어지는 각 방향으로 하나씩 두 개의 복제 포크만 있습니다. 큰 게놈을 가진 유기체에서 DNA 복제는 단일 기원 지점에서 수행되는 것이 아니라 여러 뚜렷하고 국소적인 복제 분기점에서 수행됩니다.

DNA replication stress and fork stalling can disrupt the copying of a cell’s genome. The replication fork is the Y-shaped site where DNA strands separate and new complementary strands are made at the same time. This linked unwinding and synthesis helps cells duplicate DNA efficiently and with fewer errors.

In organisms with small circular DNA, such as E. coli, replication often begins at a single origin. That setup creates two replication forks that move in opposite directions from the opening site. In organisms with large genomes, DNA copying starts at many local replication forks instead of one origin.

Smooth fork movement is needed for complete DNA replication and genome stability. When a fork slows or stops, the cell experiences replication stress. This stress can lead to genomic instability, which means more harmful mutations and other DNA changes. Genomic instability is also a hallmark of diseases such as cancer.

Several problems can stall a replication fork. Hydroxyurea is one example. It reduces the pool of nucleotides needed to build the daughter strand. DNA lesions, collisions between a replication fork and a transcription complex, and defects in DNA replication enzymes can also block fork progress.

Cells have repair systems that can restart stalled forks. S-phase checkpoints prevent cells from entering mitosis until DNA repair is finished. Fork repriming can also restart DNA synthesis by skipping over a DNA lesion or other block. Even so, some stalled forks cannot be restarted, and fork collapse stops DNA replication.
